      Abcam’s anti-Influenza virus B IgA Human in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the accurate qualitative measurement of IgA class antibodies against Influenza virus B in Human serum and plasma.

      A 96-well plate has been precoated with Influenza virus B antigens to bind cognate antibodies. Controls or test samples are added to the wells and incubated. Following washing, a horseradish peroxidase (HRP) labelled anti-Human IgA conjugate is added to the wells, which binds to the immobilized Influenza virus B-specific antibodies. TMB is then catalyzed by the HRP to produce a blue color product that changes to yellow after adding an acidic stop solution. The density of yellow coloration is directly proportional to the amount of Influenza virus B IgA sample captured in plate.

    Dear Abcam, Thanks for your reply. We intend to monitor anit-flu serum IgA from people who are vaccined with t rivalent flu vaccine, which virus strains a re A/California/7/2009 (H1N1), A/Victoria/210/2009 (H3N2) and B/Brisbane/60/2008. Here are our further inquiry. 1. Are ab108743 and ab108744 suitable for detecting our serum sample? Since the coated antigen seems not exactly the same as our vaccine strain. 2. Could we detect IgA from nasal swab sample with your ELISA kit? Do you have suggested sample preparation method for nsasal swab sample ? Thanks for your time. Best regard,

    Answer

    Thank you for your patience. Unfortunately, these kits have not been tested with the virus strains you are using and also not with nasal swaps and we do not know if this would work. I also have an additional concern: I understand you want to detect IgA antibodies which are synthesized as response to a vaccination with Influenza antigens. We have no data or literature about that but is very unlikely that the body will create IgA antibodies. This kind of antibody is a response of the mucosa to contact with a pathogen. In case of a vaccination there is no mucosa contact and hence I believe the production of IgA antibodies is improbable unless the vaccination is being administered intra nasally. I am sorry that I could not provide you with a positive answer on this occasion and hope this information is nevertheless helpful.

    Dear Abcam, We are intrested in your 2 products, Influenza Virus A IgA Human ELISA Kit (ab108743) and Influenza Virus B IgA Human ELISA Kit (ab108744). However, we have two issues for you. 1. Can you tell us which flu antigen is coated on the strip? Whole influenza virus, splitted flu antigen or influenza HA protein only or other any form of flu antigen? 2. Can you tell me what immunogen are postive control and cut-off control serum against to, ex. whole flu virus, flu HA only or other epitope? We are intended to analyse influenza split vaccine immunized human serum and we concern about the vaccine might just contain HA antigen. By the way, do you sell human anti-influenza HA IgA serum which can be used for standard control and quantitation standard curve? Thanks for your time. Best regards,

    Thank you for your inquiry. Ploease see the answer to your questions below: 1. For Influenza A IgA ELISA, ab108743, the wells are coated with purified Influenza A grade 2 antigens, strain Texas 1/77 (H3N2). For Influenza B IgA ELISA, ab108744, the wells are coated with purified Influenza B grade 2 antigens, strain Honk Kong 5/72. I can confirm that this ia a mixture of antigens and not only HA antigen. 2. I can confirm that the controls and cut-off control consists of positive human serum with preservative. Unfortunately, we do not sell human anti-influenza HA IgA serum at the moment. But I believe this is not necessary with this kit, since the control already consists of positive human serum. I hope this information is helpful and wish you good luck with your research.
