    The ELISPOT assay is designed to enumerate cytokine producing cells in a single cell suspension. This method has the advantage of requiring a minimum of in-vitro manipulations allowing cytokine production analysis as close as possible to in-vivo conditions in a highly specific way. This technique is designed to determine the frequency of cytokine producing cells under a given stimulation, and the follow-up of such frequency during a treatment and/or a pathological state. Elispot assay constitutes an ideal tool in the TH1 / TH2 response, vaccine development, viral infection monitoring and treatment, oncology, infectious diseases, autoimmune diseases and transplantation.

    This Elispot assay is based on sandwich immuno-enzyme technology. Cell secreted cytokines or soluble molecules are captured by coated antibodies avoiding diffusion in supernatant, protease degradation or binding on soluble membrane receptors. After cell removal, the captured cytokines are revealed by tracer antibodies and appropriate conjugates.

    The dual colour Elispot allows you to monitor the production of two cytokines simultaneously in the same well.

    IFN-gamma is produced by lymphocytes activated by specific antigens or mitogens. IFN-gamma, in addition to having antiviral activity, has important immunoregulatory functions. It is a potent activator of macrophages, it has antiproliferative effects on transformed cells and it can potentiate the antiviral and antitumor effects of the type I interferons.

      I would suggest reading the following protocol for ELISPOT assay. https://www.abcam.com/index.html?pageconfig=resource&rid=11456

      ELISPOT assay is used to detect interleukins or similar proteins that are secreted by live cells on membrane, which means when you infect the PBMC with antigen (tetanus toxin) then these cells will be secreting interferon's and interleukins as you are expecting to see. So you would need antibodies against interleukins rather than anti tetanus antibodies.

      Please also note that for positive control you would need is the protein that you should use to induce the known form of interleukin. So you should be searching for a protein as positive control.

      Now secondly you are interested in detecting tetanus in your blood samples for which ELISPOT is not the suitable techniques. The technique you would be looking for is indirect ELISA or sandwich ELISA. In these applications you would need anti tetanus antibodies. The antibodies will tell you how much toxin is present in your samples qualitatively as well as quantitatively.

      Now please clarify

      - are you looking for anti tetanus antibodies for ELISA?
      - Are you looking for protein to be used as positive control which induce production of interleukins in ELISPOT?
      - Are you looking for anti interleukin antibodies for ELISPOT assay?
