    Abcam’s IP-10 Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human IP-10 in serum, plasma, and cell culture supernatants.

    This assay employs an antibody specific for Human IP-10 coated on a 96-well plate. Standards and samples are pipetted into the wells and IP-10 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human IP-10 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of IP-10 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

CXCL10 secretion by macrophages

Excellent Excellent 5/5 (Ease of Use)
CXCL10 secretion my human primary macrophages in culture for 3 days.
Human monocytes isolated by negative selection.
CXCL10 secretion by the M1 polarized macrophages is observed.
The kit worked perfectly well, no dilution necessary, the assay was performed in 5.30hr.

I have contacted the lab who confirmed that it is not recommended to load less than 100 ul of sample per well as this may affect the quality of the results. For the best results, the protocol should be followed exactly.

Thank you for your inquiry. Unfortunately, there is no cross reactivity data available testing cynomolgus with the Human IP-10 ELISA kit (ab100579). However, I would expect the homology between cynomolgus and human to be quite high so I think it would definitely be worth a shot. If it helps I can release that the capture antibody is a monoclonal mouse IgG1 and the detection antibody is a polyclonal goat IgG. Please let me know if you have any questions or comments.
