    LYVE1 in vitro SimpleStep ELISA® (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of LYVE1 protein in human serum, plasmas, and urine.

    The SimpleStep ELISA® employs an affinity tag labeled capture antibody and a reporter conjugated detector antibody which immunocapture the sample analyte in solution. This entire complex (capture antibody/analyte/detector antibody) is in turn immobilized via immunoaffinity of an anti-tag antibody coating the well. To perform the assay, samples or standards are added to the wells, followed by the antibody mix. After incubation, the wells are washed to remove unbound material. TMB Development Solution is added and during incubation is catalyzed by HRP, generating blue coloration. This reaction is then stopped by addition of Stop Solution completing any color change from blue to yellow. Signal is generated proportionally to the amount of bound analyte and the intensity is measured at 450 nm. Optionally, instead of the endpoint reading, development of TMB can be recorded kinetically at 600 nm.

    Lymphatic Vessel Endothelial Hyaluronic Acid Receptor 1 (LYVE1), also known as Cell surface retention sequence-binding protein 1, Extracellular link domain-containing protein 1, and hyaluronic acid receptor is a CD44 homolog protein encoded by the LYVE1 gene. LYVE1 is a type I integral membrane glycoprotein that acts as a receptor to both soluble and immobilized hyaluronan and is located primarily on lymphatic endothelial cells. LYVE1 forms a disulfide- linked homodimer and interacts with PDGFB and IGFBP3.

  • The LYVE1 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.
  • The concentrations of LYVE1 were measured in duplicates, interpolated from the LYVE1 standard curves and corrected for sample dilution. Undiluted samples are as follows: plasma (EDTA) 1:500, plasma (citrate) 1:500, and plasma (heparin) 1:500. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean LYVE1 concentration was determined to be 375 ng/mL in plasma (EDTA), 591 ng/mL in plasma (citrate) and 372 ng/mL in plasma (Heparin).
  • The concentrations of LYVE1 were measured in duplicates, interpolated from the LYVE1 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 1:500, urine 1:20. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean LYVE1 concentration was determined to be 486 ng/mL in serum and 33 ng/mL in urine.
