  • Product overview

Abcam’s MMP3 Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human MMP3 pro and active forms in serum, plasma, and cell culture supernatants. (Collect plasma using heparin as an anticoagulant. EDTA and Citrate are not recommended).

This assay employs an antibody specific for Human MMP3 coated on a 96- well plate. Standards and samples are pipetted into the wells and MMP3 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human MMP3 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of MMP3 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Question
Answer

Thank you for your email.

I am happy to process the replacement. Could you please confirm how many kits (ab100607) your customer ordered originally?

Thank you.

Question

Please see below customer's answers:

1.) When has the kit been used and when have the components been reconstituted? How has the kit before and after reconstitution been stored?

The kit was reconstituted and used on the same day, May 21st 2012 (together with the MMP-1 kit). Both before and after reconstitution they were kept at 4 deg C

2.) Did you have any troubles reconstituting the standard?

No. We subsequently tested the standard dilutions by dot blot with another anti-MMP3 antibody an obtained a reasonable signal.

3.) Have you any comment on which component might be faulty? Have you, for example, tested whether the adding some of the HRP conjugated antibody to an aliquot of the substrate itself would give a signal?

We have no idea for “ faulty component” but we can mention that the substrate was OK since we used the pooled substrates from kit MMP-3 and MMP-1 for both assays - in MMP1 plate we got a nice standard curve. We have indication that standard was OK since we got reaction with another batch of anti MMP3 antibody in a different method. We did not test the HRP antibody.

4.) Just for our records, what kind of samples have you used?

Our samples are skin surface washes of patients suffering from dermatitis and psoriasis (a small vessel with PBS is attached to skin for 30 min). Lesional skin from such patients is known to over-express these proteins. And indeed, in a previous group of patients tested by dot blot (and by using different antibodies) we obtained the expected results.

Please advise.

Thanks in advance.

Answer

Thank you very much for confirming all these details. This is indeed very useful for us.
It looks like the customer has received a kit with a faulty HRP / HRP conjugate. We will therefore be happy to replace the kit for the customer. Please let us know if the customer would accept a replacement kit or if he would prefer a credit note. I apologize for all the inconvenience caused.
I look forward hearing back from you.

Answer

rmmarize the problem. I am sorry to hear that this kit is not providing satisfactory results.
We have not yet received any negative feedback and I am not able yet to tell where the problem might is coming from. I would therefore appreciate if you can confirm some further details:
1.) When has the kit been used and when have the components been reconstituted? How has the kit before and after reconstitution been stored?
2.) Did the customer had any troubles reconstituting the standard?
3.) Has the customer any comment on which component might be faulty? Has he for example tested whether the adding some of the HRP conjugated antibody to an aliquot of the substrat itself would give a signal?
4.) Just for our records, what kind of samples has the customer used?
In the event that a product is not functioning in the species and applications cited on the product datasheet (and the problem has been reported within 6 months of purchase), we would be pleased to provide a free of charge replacement, credit note, or refund.
I hope this information is helpful, and I thank you for your cooperation.

Answer

Thank you for your interest in the MMP-3 and IL23 kits.

The volume of sample required for each test is 100ul per well of the assay plate.

Regarding the 100% discount, I want to be sure you understand the offer, which is also described at the following link:

https://www.abcam.com/index.html?pageconfig=resource&rid=11998&viapagetrap=collaborationdiscount

The discount is not applied to the purchase of the kits. The kits are purchased full-price after we issue you a code. After you test the kits on the monkey serum, you then send Abcam a review of your results (with the code ID in the review), via our Abreview form. ( Please see www.abcam.com/abreviews).

After we receive your reviews, the code is then "active" and can be redeemed for a free kit. Or, it can be used to receive a 20% discount on a primary antibody or a protein.

In addition, 440 points will be awarded for the first review published for ELISA products. Please see https://www.abcam.com/index.html?pageconfig=resource&rid=14713 for more details.

These offers are available through June.

Please let me know if you are still interested, and I will send you the codes, which are necessary before you place your order.

Abreview for MMP3 Human ELISA Kit

Good Good 4/5 (Ease of Use)
Abreviews

Sample: Human Cell (Skin fibroblasts)
Specification: Skin fibroblasts
Amount of Sample: 200 µg
Type of Kit Used: ELISA
Positive control: Included in kit as part of standard curve.
Did you use the Abcam Kit protocol?: Yes

Additional data
Additional Notes: Incubation times are a little longer than in some other kits, which must be remembered when planning multiple experiments, but the data generated is of a better quality overall.

The graph shows the differing levels of MMP3 released by primary fibroblasts exposed to several different active ingredients dissolved in media for several days prior to media collection and analysis.

(Sorry I cant say what the actives actually are due to business nature of the work).

Answer

Thank you for your interest in our kits. I will send the product inserts first thing tomorrow morning, EST. The kits will be compatible with the automated ELISA machine, if the automated set-up approximates the conditions specified in the kit protocols. We provide a discount for orders of 5 or more kits. I will have our customer service department prepare a proforma. I do not anticipate any issues with the age of the sera, if they have been stored at a consistent temperature of -20C or less. You may want to conduct a trial before committing to a large run, however.
