  • Product overview

Abcam’s Osteopontin Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human Osteopontin in serum, plasma, and cell culture supernatants.

This assay employs an antibody specific for Human Osteopontin coated on a 96-well plate. Standards and samples are pipetted into the wells and Osteopontin present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human Osteopontin antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Osteopontin bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Though we have not tested the protein ab81549 (https://www.abcam.com/osteopontin-protein-active-ab81549.html) with ab100618. However it is most likely to work.
I hope this information is helpful to you. Please do not hesitate to contact us if you need any more advice or information.

Thank you for your enquiry.

I can confirm that the suggested dilution for normal serum/plasma when testing with this kit is 3-100 fold.

Please note that this will depend on expression level in the samples you have and so this is a guideline only. This may require some further optimization by the end user.

I hope this will be helpful to you. If you have any further questions, please do not hesitate to contact us.

I am happy to confirm that we offer ELISA kits for Osteopontin:

ab100618 (Osteopontin Human ELISA Kit)

https://www.abcam.com/index.html?datasheet=100618 (or use the following: https://www.abcam.com/index.html?datasheet=100618).

ab100734 (Osteopontin Mouse ELISA Kit)

https://www.abcam.com/index.html?datasheet=100734 (or use the following: https://www.abcam.com/index.html?datasheet=100734).

Unfortunately, we do not have antibodies that are tested and guaranteed to only recognize a single specific isoform of Osteopontin in our catalog.

I hope this information ishelpful and wish you good luck for your experiments.

Thank you for contacting us.

We received the following explanation from the lab:

The full length Osteopontin protein (aa 17-314) was used as the immunogen for the antibodies used in thekit ab100618. And as epitope mapping was not performed, we do not know exactly where the antibodies are binding. Therefore it is not known whether this kit will recognize the cleaved forms (or consequently if it is able to differentiate between the different forms).

I hope this information is of some help to you. Please do not hesitate to contact us if you need any more advice or information.
