    PD-L2 in vitro SimpleStep ELISA® (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of PD-L2 protein in human serum, plasma, urine, milk, saliva, and cell culture supernatant.


    The SimpleStep ELISA® employs an affinity tag labeled capture antibody and a reporter conjugated detector antibody which immunocapture the sample analyte in solution. This entire complex (capture antibody/analyte/detector antibody) is in turn immobilized via immunoaffinity of an anti-tag antibody coating the well. To perform the assay, samples or standards are added to the wells, followed by the antibody mix. After incubation, the wells are washed to remove unbound material. TMB substrate is added and during incubation is catalyzed by HRP, generating blue coloration. This reaction is then stopped by addition of Stop Solution completing any color change from blue to yellow. Signal is generated proportionally to the amount of bound analyte and the intensity is measured at 450 nm. Optionally, instead of the endpoint reading, development of TMB can be recorded kinetically at 600 nm.


    PD-L2 (PDCD1LG2), Programmed cell death 1 ligand 2, has overlapping functions with PD-L1. PD-L1 suppresses the immune system during pregnancy, tissue allografts and autoimmune diseases. PD-L1 bind to PD-1 and CTLA-4 inhibiting the proliferation of activated T-cells. PD-L2 functions the same, but seems to exclusively bind PD-1. PD-L1 and PD-L2 cross compete for PD-1 binding.

  • The concentrations of PD-L2 were measured in duplicates, interpolated from the PD-L2 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 5%, plasma (citrate) 10%, plasma (EDTA) 5%, and plasma (heparin) 5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean PD-L2 concentration was determined to be 9.7 ng/mL in serum, 11.1 ng/mL in plasma (citrate), 7.6 ng/mL in plasma (EDTA) and 8.5 ng/mL in plasma (heparin).

  • Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean PD-L2 concentration was determined to be 10.3 ng/mL with a range of 7.5 – 15.0 ng/mL.

  • The concentrations of PD-L2 were measured in duplicates, interpolated from the PD-L2 standard curves and corrected for sample dilution. Undiluted samples are as follows: urine 50% and milk 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean PD-L2 concentration was determined to be 2172 pg/mL in urine and 151 pg/mL in milk.

  • The concentrations of PD-L2 were measured in duplicates, interpolated from the PD-L2 standard curves and corrected for sample dilution. Undiluted samples are as follows: saliva 50% and cell culture media 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2).

  • The concentrations of PD-L2 were measured in duplicates, interpolated from the PD-L2 standard curves and corrected for sample dilution. Undiluted samples are as follows: unstimulated 100% and stimulated 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean PD-L2 concentration was determined to be 32 pg/mL in stimulated PBMC cell culture supernatant and undetectable in unstimulated and media controls.
