    Abcam’s Perforin (PRF1) Human in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of Perforin in supernatant and buffered solutions.

    A monoclonal antibody specific for Perforin has been coated onto the wells of the microtiter strips provided. Samples, including standards of known Perforin concentrations, control specimens or unknowns are pipetted into these wells. During the first incubation, the standards or samples and a biotinylated monoclonal antibody specific for Perforin are simultaneously incubated. After washing, the enzyme Streptavidin-HRP, that binds the biotinylated antibody is added, incubated and washed. A TMB substrate solution is added which acts on the bound enzyme to induce a colored reaction product. The intensity of this colored product is directly proportional to the concentration of Perforin present in the samples. 

    This kit will recognize both endogenous and recombinant Human Perforin.

    Please note this kit is not suitable for use with serum and plasma.

    Plays a key role in secretory granule-dependent cell death, and in defense against virus-infected or neoplastic cells. Plays an important role in killing other cells that are recognized as non-self by the immune system, e.g. in transplant rejection or some forms of autoimmune disease. Can insert into the membrane of target cells in its calcium-bound form, oligomerize and form large pores. Promotes cytolysis and apoptosis of target cells by facilitating the uptake of cytotoxic granzymes.

May be taken up via endocytosis involving clathrin-coated vesicles and accumulate in a first time in large early endosomes.

Regarding Perforin ELISA Kit, standard batch contains 3060 pg/vial. However, the following batch contains 1820 pg/vial. I hope this will be useful for you.

Thank you for your enquiry. The standard for both kits is lyophilised, thus it is not possible to provide the concentration in the vial. However, there will be a batch number-specific information/instruction printed on the label of the vial. This will guide the customer how to resuspend the vial and the final concentration after reconstitution will be 2000 pg/ml.
