      Abcam’s PLGF Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human PLGF in serum, plasma, and cell culture supernatants.

      This assay employs an antibody specific for Human PLGF coated on a 96-well plate. Standards and samples are pipetted into the wells and PLGF present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human PLGF antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of PGF bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

      Growth factor active in angiogenesis and endothelial cell growth, stimulating their proliferation and migration. It binds to the receptor FLT1/VEGFR-1. Isoform PlGF-2 binds NRP1/neuropilin-1 and NRP2/neuropilin-2 in a heparin-dependent manner.
    • Tissue specificity

      While the three isoforms are present in most placental tissues, PlGF-2 is specific to early (8 week) placenta and only PlGF-1 is found in the colon and mammary carcinomas.
    • Sequence similarities

      Belongs to the PDGF/VEGF growth factor family.
    • Domain

      Isoform PlGF-2 contains a basic insert which acts as a cell retention signal.

      N-glycosylated.

      Secreted. The three isoforms are secreted but PlGF-2 appears to remain cell attached unless released by heparin.

      Inquiry: The protocol doesn't give any indications about sample diluition. I tried the 1:2 diluition, but it's gone on saturation. Same thing for ab119613 and ab100629. Please tell me how to go on. Thank you very much.

      Optimal dilution of the sample will usually need to be optimized by the end user. In this case, I would appreciate if you could let me know what type of samples you are using, and I can find out some recommended dilutions for you for these kits.
