    - First try to dissolve a small amount of peptide in either water or buffer. The more charged residues on a peptide, the more soluble it is in aqueous solutions.
    - If the peptide doesn’t dissolve try an organic solvent e.g. DMSO, then dilute using water or buffer.
    - Consider that any solvent used must be compatible with your assay. If a peptide does not dissolve and you need to recover it, lyophilise to remove the solvent.
    - Gentle warming and sonication can effectively aid peptide solubilisation. If the solution is cloudy or has gelled the peptide may be in suspension rather than solubilised.
    - Peptides containing cysteine are easily oxidised, so should be prepared in solution just prior to use.

    Has both folate hydrolase and N-acetylated-alpha-linked-acidic dipeptidase (NAALADase) activity. Has a preference for tri-alpha-glutamate peptides. In the intestine, required for the uptake of folate. In the brain, modulates excitatory neurotransmission through the hydrolysis of the neuropeptide, N-aceylaspartylglutamate (NAAG), thereby releasing glutamate. Isoform PSM-4 and isoform PSM-5 would appear to be physiologically irrelevant. Involved in prostate tumor progression.
    Also exhibits a dipeptidyl-peptidase IV type activity. In vitro, cleaves Gly-Pro-AMC.
  • Tissue specificity

    Highly expressed in prostate epithelium. Detected in urinary bladder, kidney, testis, ovary, fallopian tube, breast, adrenal gland, liver, esophagus, stomach, small intestine, colon and brain (at protein level). Detected in the small intestine, brain, kidney, liver, spleen, colon, trachea, spinal cord and the capillary endothelium of a variety of tumors. Expressed specifically in jejunum brush border membranes. In the brain, highly expressed in the ventral striatum and brain stem. Also expressed in fetal liver and kidney. Isoform PSMA' is the most abundant form in normal prostate. Isoform PSMA-1 is the most abundant form in primary prostate tumors. Isoform PSMA-2 is also found in normal prostate as well as in brain and liver. Isoform PSMA-9 is specifically expressed in prostate cancer.

    The NAALADase activity is found in the central region, the dipeptidyl peptidase IV type activity in the C-terminal.

Question

Further to the earlier enquiry, I have a few additional questions regarding your PSMA peptide. You have stated that the PSMA peptide (ab41442) is 17 amino acids in length; however, the datasheet provided states the length as 750 amino acids - is this the length of the whole human PSMA from which the peptide was derived?

I would also like to ask what the differences are between ab41442 and ab97412; and if there are any differences in storage buffer with the PSMA peptide (ab97412) and the PSMA protein (ab117471).

Thank you for your reply.

You are correct in thinking that the description of the "protein length" described for ab41442 refers to the full length of the PSMA protein from which the peptide is taken. The peptide is described as

"Synthetic peptide derived from within residues 1 - 100 of Human PSMA.(Note: the amino acid sequence is proprietary)This peptide was used as an immunogen for https://www.abcam.com/ab41034.html."

The protein ab117471 is in a slightly different buffer to ab41442. It is provided in a Tris buffer (pH 7.5) with 0.31% Glutathione.

I hope this information has been of help. If you could describe what you are hoping to use the protein for I can advise further as to whether either of these products, or anything else from our catalogue, might be suitable.

The PSMA peptide (ab41442) is a 17 amino acid long peptide derived from within the first 100 residues of human PSMA. It was used as the immunogen to produce the anti-PSMA anditbody ab41034. Please find below the answers to your questions:

Stability and shelf life of the peptide.



We would recommend storing the peptide as directed on the datasheet of ab41442:

Upon delivery aliquot and store at -20°C or -80°C. Avoid repeated freeze / thaw cycles.

We do not have set expiration dates for our products. Most peptides are stable and can last for anywhere from a few months to several years if stored properly, so we strongly recommend that you follow the storage instructions on the datasheet for the product you purchase. These conditions will vary among our products, therefore, it is important to verify the storage conditions for each of our products when you receive them. We guarantee all of our products to work for at least 120 days from the date of purchase when stored correctly.


Formulations and peptide sequences if available.



The peptide ab41442 is provided at a concentration of 1 mg/ml in a HEPES buffer (pH 6.75) with EDTA, sodium chloride and Tween (0.001%)

I was just wondering if you might be able to tell me why you chose to immunize with the 16 AA PSMA peptide over the 18 AA PSMA peptide or the PSMA protein?

I am planning on immunizing a chicken and a rabbit with one of your products in the next 3 weeks and I was just curious as to why you chose that one to immunize with to make ab41034.

Indeed, ab41442 was used as an immunogen to make one of our PSMA antibodies, ab41034. However, ab97421 has also been used as an immunogen to make another one of our PSMA antibodies, ab71720. Therefore, with regards to your question both peptides are equally capable of being able to produce antibodies against PSMA.
