    PTH in vitro SimpleStep ELISA® (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of PTH protein in Human serum, plasma and cell culture media.


    The SimpleStep ELISA® employs an affinity tag labeled capture antibody and a reporter conjugated detector antibody which immunocapture the sample analyte in solution. This entire complex (capture antibody/analyte/detector antibody) is in turn immobilized via immunoaffinity of an anti-tag antibody coating the well. To perform the assay, samples or standards are added to the wells, followed by the antibody mix. After incubation, the wells are washed to remove unbound material. TMB substrate is added and during incubation is catalyzed by HRP, generating blue coloration. This reaction is then stopped by addition of Stop Solution completing any color change from blue to yellow. Signal is generated proportionally to the amount of bound analyte and the intensity is measured at 450 nm. Optionally, instead of the endpoint reading, development of TMB can be recorded kinetically at 600 nm.


    Parathyroid hormone (PTH) also called parathormone or parathyrin, is secreted by the parathyroid gland. It has known functions in bone remodeling processes, bone tissue reabsorption and bone reconstruction. PTH is secreted in response to low blood serum calcium (Ca2+) levels. PTH elevates calcium levels by dissolving the salts in bone and preventing their renal excretion. PTH also stimulates [1-14C]-2-deoxy-D-glucose (2DG) transport and glycogen synthesis in osteoblastic cells.  Human PTH shares 70% and 73% sequence homology with mouse and rat DNA.

  • Involvement in disease

    Defects in PTH are a cause of familial isolated hypoparathyroidism (FIH) [MIM:146200]; also called autosomal dominant hypoparathyroidism or autosomal dominant hypocalcemia. FIH is characterized by hypocalcemia and hyperphosphatemia due to inadequate secretion of parathyroid hormone. Symptoms are seizures, tetany and cramps. FIH exist both as autosomal dominant and recessive forms of hypoparathyroidism.

  • The concentrations of PTH were measured in duplicate, interpolated from the PTH standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 50%, plasma (citrate) 50%, plasma (EDTA) 50%, plasma (heparin) 50% and 10F RPMI media 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean PTH concentration was determined to be 262.33 pg/mL in serum, 258.51 pg/mL in plasma (citrate) and 264.22 pg/mL in plasma (EDTA), 249.32 pg/mL in plasma (heparin), and 300.25 pg/mL in 10F RPMI media 100%.
