    The Human S100A12 Enzyme-Linked Immunosorbent Assay (ELISA) kit (ab213822) is designed for the quantitative measurement of Human S100A12 in cell culture supernatants, cell lysates, tissue homogenates, serum and plasma (heparin).

    The ELISA kit is based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for S100A12 has been pre-coated onto 96-well plates. Standards and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for S100A12 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. HRP substrate TMB is used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Human S100A12 amount of sample captured in plate.

    S100 calcium-binding protein A12 (S100A12) is a protein that in humans is encoded by the S100A12 gene. It is also known as calgranulin C. By direct R-banding FISH, the S100A12 gene is mapped to chromosome 1q21.2-q22. The protein encoded by this gene is a member of the S100 family of proteins containing 2 EF-hand calcium-binding motifs. S100 proteins are localized in the cytoplasm and/or nucleus of a wide range of cells, and involved in the regulation of a number of cellular processes such as cell cycle progression and differentiation. S100 genes include at least 13 members which are located as a cluster on chromosome 1q21. And this protein is proposed to be involved in specific calcium-dependent signal transduction pathways and its regulatory effect on cytoskeletal components may modulate various neutrophil activities.

    Calcitermin possesses antifungal activity against C.albicans and is also active against E.coli and P.aeruginosa but not L.monocytogenes and S.aureus. Binds calcium, zinc and copper. Presence of zinc increases the affinity for calcium. Plays an important role in the inflammatory response. Interaction with AGER on endothelium, mononuclear phagocytes, and lymphocytes triggers cellular activation, with generation of key proinflammatory mediators.
