    Abcam’s Soluble TNF Receptor II ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human Soluble TNF Receptor II in serum, plasma and cell culture supernatants.

    This assay employs an antibody specific for Human Soluble TNF Receptor II coated on a 96-well plate. Standards and samples are pipetted into the wells and Soluble TNF Receptor II present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human Soluble TNF Receptor II antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Soluble TNF Receptor II bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

    TNFRII (TNF Receptor II) is a member of the TNFR family of transmembrane proteins, and is expressed in immune cells and certain endothelial cells. It is a high affinity receptor for TNF alpha but manifests a lower affinity to TNF beta. Signaling through this receptor regulates various biological processes including cell proliferation, differentiation, apoptosis, lipid metabolism, coagulation, and neurotransmission. TNFRII consists of a 240 amino acid residue extracellular ligand binding portion, a 27 amino acid residue transmembrane segment and a 173 amino acid residue cytoplasmic domain. Soluble TNFRII (Soluble TNF Receptor II) is a 268 amino acid isoform containing the extracellular ligand-binding portion of TNFRII. It binds to TNF alpha and is capable of inhibiting TNF alpha induced activities by acting as a decoy receptor.

I can confirm that this kit (Soluble human TNF Receptor II) is suitable for detecting Etanarcept which is an alternative name for TBPII. We have not specifically tested with these drugs. As long as your samples have detectable levels of TBPII and the samples are from cell culture supernatant, urine, serum, or from plasma this kit should work.

I would suggest reading through the Protocol Booklets for each of these kits provided on the product datasheets. These documents contain specific information about the sample types, sample preparations, protocol, instruments and equipment required for the assays. Whole blood samples are not used to test these kits mainly serum or plasma instead.
