Human Sortilin 1 ELISA kit (ab277074) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human Sortilin 1 protein in human tissue extract samples. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Sortilin 1 with 261.7 pg/mL sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Sortilin 1 is a receptor found in the Golgi and is involved in protein sorting and transport. Sortilin 1 is ubiquitously expressed and abundant in neurons, where it is involved in neuronal development and apoptosis. Sortilin 1 is also involved in lipid metabolism and has been suggested as a clinical marker for coronary artery disease. Sortilin 1 has been implicated in atherosclerosis, Alzheimer’s disease, and cancer.

FunctionFunctions as a sorting receptor in the Golgi compartment and as a clearance receptor on the cell surface. Required for protein transport from the Golgi apparatus to the lysosomes by a pathway that is independent of the mannose-6-phosphate receptor (M6PR). Also required for protein transport from the Golgi apparatus to the endosomes. Promotes neuronal apoptosis by mediating endocytosis of the proapoptotic precursor forms of BDNF (proBDNF) and NGFB (proNGFB). Also acts as a receptor for neurotensin. May promote mineralization of the extracellular matrix during osteogenic differentiation by scavenging extracellular LPL. Probably required in adipocytes for the formation of specialized storage vesicles containing the glucose transporter SLC2A4/GLUT4 (GLUT4 storage vesicles, or GSVs). These vesicles provide a stable pool of SLC2A4 and confer increased responsiveness to insulin. May also mediate transport from the endoplasmic reticulum to the Golgi.

Involvement in diseaseNote=A common polymorphism located in a non-coding region between CELSR2 and PSRC1 alters a CEBP transcription factor binding site and is responsible for changes in hepatic expression of SORT1. Altered SORT1 expression in liver affects low density lipoprotein cholesterol levels in plasma and is associated with susceptibility to myocardial infarction.

The Sortilin 1 standard curve was prepared as described in Section 10. Raw data values are shown in the table. Background-subtracted data values (mean +/- SD) are graphed.
The concentrations of Sortilin 1 were measured in duplicate and interpolated from the Sortilin 1 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Sortilin 1 concentration was determined to be 20,619 pg/ml in cortex homogenate extract.
