      Abcam’s TIMP1 Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human TIMP1 in cell lysates and tissue lysates.


      This assay employs an antibody specific for Human TIMP1 coated on a 96-well plate. Standards and samples are pipetted into the wells and TIMP1 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human TIMP1 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of TIMP1 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

      Complexes with metalloproteinases (such as collagenases) and irreversibly inactivates them by binding to their catalytic zinc cofactor. Also mediates erythropoiesis in vitro; but, unlike IL-3, it is species-specific, stimulating the growth and differentiation of only human and murine erythroid progenitors. Known to act on MMP-1, MMP-2, MMP-3, MMP-7, MMP-8, MMP-9, MMP-10, MMP-11, MMP-12, MMP-13 and MMP-16. Does not act on MMP-14.

      Thank you very much for contacting. I apologize for the delay as we were conducting some tests.

      We have also seen gloopy things in the Assay buffer which first seen as like fungal growth however when we warmed the buffer at room temperature which is the working temperature of buffer; the precipitates were vanished.

      Because the assay diluent contains some proteins and solutes so it would not be uncommon to see cloudiness or floating unsolubilized protein aggregates (especially after storing at low temperatures and/or for a long period of time). We recommend bringing the solution up to room temperature, mixing it thoroughly and diluting to the 1X working concentration which will help clarifying the solution and solubilising any protein aggregates or precipitates. Centrifuging and filtering can also help.

      Question

      I don't know if you are the correct person to contact, if not please could you forward my email to the relevant department.
      I purchased the four ELISA kits listed below on the 23rd March and received them just before Easter (PO 68241150). They have been stored in their boxes, unopened as recommended at 2-4oC since then. When my student started using the TIMP1 kit yesterday, we discovered that in the assay diluent buffer bottle there was a large amount of filamentous or fibrous white substance floating in the buffer. I looked at it under the microscope and it could be fungal contamination but I could not say for certain. It did not appear to be something that had precipitated out of the buffer (and there was no mention in the kit to watch for any kind of precipitation, which was in the manual for the wash buffer). When we checked the other kits which were unopened, we discovered that 3 out of the 4 kits had this substance in the assay diluent buffer to varying amounts. Only the MMP9 kit did not have any in its assay diluent buffer.
      Please could you advise us what to do about this and whether we could be sent a new batch of assay diluent buffer for the 4 kits?
