    Human Transferrin SimpleStep ELISA® kit has been re-developed. We have identified new recombinant monoclonal antibodies to provide improved performance and consistency. This version will be discontinued April 2020 unless inventory is depleted beforehand.


    Transferrin in vitro SimpleStep ELISA® (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of Transferrin protein in human serum, plasma and cell culture supernatant samples.


    The SimpleStep ELISA® employs an affinity tag labeled capture antibody and a reporter conjugated detector antibody which immunocapture the sample analyte in solution. This entire complex (capture antibody/analyte/detector antibody) is in turn immobilized via immunoaffinity of an anti-tag antibody coating the well. To perform the assay, samples or standards are added to the wells, followed by the antibody mix. After incubation, the wells are washed to remove unbound material. TMB substrate is added and during incubation is catalyzed by HRP, generating blue coloration. This reaction is then stopped by addition of Stop Solution completing any color change from blue to yellow. Signal is generated proportionally to the amount of bound analyte and the intensity is measured at 450 nm. Optionally, instead of the endpoint reading, development of TMB can be recorded kinetically at 600 nm.

    Transferrins are iron binding transport proteins which can bind two Fe3+ ions in association with the binding of an anion, usually bicarbonate. It is responsible for the transport of iron from sites of absorption and heme degradation to those of storage and utilization. Serum transferrin may also have a further role in stimulating cell proliferation.

Yes, the capture and detector antibody recognize human Transferrin.

1) The antibodies in this kit have been raised against native transferrin and unfortunately we do not know precise epitopes. A monoclonal capture antibody and a polyclonal detector antibody is provided. Therefore we do not know if the mutations generated will be recognized.

2 and 3) We have not tested the effects of bicarbonate, oxycholate or iron on assay activity. The standard protein in the kit is native purified human transferrin. The kit works with this protein and with the human serum/plasma samples and has very good linearity of dilution. Therefore we expect that this antibody pair is compatible with normal levels of bicarbonate/oxycholate/iron bound to transferrin. However we do not know if abnormally high/low levels of any of these will affect the reactivity of the sample.
