WB Use a concentration of 0.25 µg/ml. Predicted molecular weight: 52 kDa. Good results were obtained when blocked with 5% non-fat dry milk in 0.05% PBS-T.
IHC-P Use a concentration of 4 - 8 µg/ml. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol.

    Adenylyltransferase that mediates the addition of adenosine 5'-monophosphate (AMP) to specific residues of target proteins. Able to inactivate Rho GTPases in vitro by adding AMP to RhoA, Rac and Cdc42. It is however unclear whether it inactivates GTPases in vivo and physiological substrates probably remain to be identified.

Just to confirm, ab104782 has been raised against a synthetic peptide immunogen "corresponding to a region within the C terminal amino acids 351-400". This peptide is a 15-20 amino acid peptide found withinamino acids 350-400, not the entire 50 amino acid region.

In your aa187-437 mutant, is the protein actually missing amino acids 187 through 437? If this is the case the the epitope recognized by ab104782 has been removed from the mutant protein the protein will no longer bind the antibody.
