    ICAM proteins are ligands for the leukocyte adhesion protein LFA-1 (integrin alpha-L/beta-2). During leukocyte trans-endothelial migration, ICAM1 engagement promotes the assembly of endothelial apical cups through ARHGEF26/SGEF and RHOG activation. In case of rhinovirus infection acts as a cellular receptor for the virus.

Reviewing this case, I would like to offer some suggestions to help optimise the results from ab33894.

-It is important to emphasize that there are at least 10 potential N-glucosylation sites along the polypeptide chain of ICAM1. As it is a glycoprotein it contains one or more covalently linked carbohydrates of various types, i.e. from monosaccharides to branched polysaccharides, including glycosylphosphatidylinositol (GPI), glycosaminoglycans (GAG), this modification will affect the MW and could lead to numerous bands at different MW depending on the sample processing.

-In cases where multiple non specific bands are present, the use of a positive control becomes indispensable to help identify the target protein. ICAM1 is highly expressed in lung, placenta and liver tissue.

- Reduce protein load from 30ug to 15ug.

-During sample processing, it is important to disrupt the disulphide bonds. I would therefore recommend increasing the reducing conditions.

-Insufficient blocking conditions could also lead to multiple bands. I would recommend increasing the blocking conditions to Milk 5% during 2 hours at RT.

- Regarding the primary antibody, I would suggest a less concentrated dilution, e.g. 1/250 or 1/500, incubating at 4C overnight.

-Increase the washing conditions. I would suggest performing 4 x 10 minutes washings.

-Run a non primary control to dismiss non specific bands coming from the secondary.
