Target

  • Function
The insulin-like growth factors, isolated from plasma, are structurally and functionally related to insulin but have a much higher growth-promoting activity. May be a physiological regulator of [1-14C]-2-deoxy-D-glucose (2DG) transport and glycogen synthesis in osteoblasts. Stimulates glucose transport in rat bone-derived osteoblastic (PyMS) cells and is effective at much lower concentrations than insulin, not only regarding glycogen and DNA synthesis but also with regard to enhancing glucose uptake.
  • Involvement in disease
Defects in IGF1 are the cause of insulin-like growth factor I deficiency (IGF1 deficiency) [MIM:608747]. IGF1 deficiency is an autosomal recessive disorder characterized by growth retardation, sensorineural deafness and mental retardation.
  • Sequence similarities
    Belongs to the insulin family.
  • Cellular localization
    Secreted.
  • Information by UniProt
  • Database links
  • Form
There are 2 isoforms produced by alternative splicing. Isoform 1 also known as: IGF-IB; Isoform 2 also known as: IGF-IA.

Question

Dear technical support team: This customer has already purchased ab36532 (Anti-IGF1 antibody), and after she conducted wb with rat sample, the result shows no band, therefore she wants to ask for you help to offer any suggestion to her, I also attached her result in this letter and her experiment step as follow: 1. Order details: Batch number: gr5099-4 Abcam product code: ab36532 Antibody storage conditions (temperature/reconstitution etc) -20oC 2. Please describe the problem (high background, wrong band size, more bands, no band etc). No band 3. On what material are you testing the antibody in WB? Species: RAT What cell line or tissue tissue- skeletal muscles-sol-EDL-gas Cell extract or Nuclear extract: tissue extract Purified protein or Recombinant protein: 3. The lysate How much protein was loaded: 30ug What lysis buffer was used: 50mM potassium phosphate,1mM EDTA,1mM DTT,0.25mM PMSF ,2% CHAPS,3M urea ,pH= 7 What protease inhibitors were used: What loading buffer was used: Phosphatase inhibitors Did you heat the samples: temperature and time: 4. Electrophoresis/Gel conditions/ Transfer conditions Reducing or non reducing gel: non reducing gel Reducing agent: Gel percentage : 12% gel Transfer conditions: (Type of membrane, Protein transfer verified): 110V 100mins 5. Blocking conditions Buffer: 5%fish (Tris、NaCl、fish gelatin) Blocking agent: milk, BSA, serum, what percentage: Incubation time: 1hr Incubation temperature: RT 6. Secondary Antibody Species: Reacts against: mouse At what dilution(s) have you tested this antibody: 1:10000 Incubation time: 1hr Wash steps: 1% fish (Tris、NaCl、fish gelatin) Fluorochrome or enzyme conjugate: Do you know whether the problems you are experiencing come from the secondary? No 8. Detection method ECl, ECl+, other detection method: ECL 9. Did you apply positive and negative controls along with the samples? Please specify. NO 10. Optimization attempts How many times have you tried the Western? 3 times Have you eliminated the possibility that any background bands could be due to the secondary antibody? (Run a “No primary” control): no. Do you obtain the same results every time e.g. are background bands always in the same place? yes What steps have you altered? Concentration of primary antibody Could you please help this customer to solve the problem? Thanks for your kindly help Best regards

Answer

Thank you for taking the time to complete our questionnaire and contact us. I am sorry to hear you have had difficulty obtaining satisfactory results from this antibody. I would like to reassure you that this antibodyis tested and covered by our guarnatee for rat and WB. In the event that a product is not functioning in the applications cited on the product data sheet (and the problem has been reported within 6 months of purchase), we will be pleased to provide a credit note, free of charge replacement or refund. The details you have kindly provided will enable us to investigate this case for you and this is also helpful in our records for monitoring of quality. Reviewing this case, I would like to offer some suggestions to help optimize the results from ab36532. I would also appreciate if you can confirm some further details: 1. I am sorry I am not able to confirm the expression level of IGF1 in skeletal protein with a literature search. Do you have any informationon this? I can suggest to consider including a positive control sample such as plasma, A549 cell line lysate orin this case. 2. Could you confirmif there is there any background staining on the blot? 3. I can suggest to consider try RIPA lysis buffer which may provide a more suitable protein preparation for this protein and epitope. 4. Could you confirm ifthe samples heated to reduce and denature? Please provide details. I can also recommend it would be beneficial to use a reducing gel. We suggest to use reducing and denaturing conditions for western blot with our antibodies, unless it recommends otherwise on the datasheet. 5. I can suggest to try BSA rather than fish gelatin to block. Changing blocking agents can sometimes improve the results. 6. I recommend to try the antibody at a higher concentration in order to increase the signal. Try 1:1000 and 1:500. 7. Is the secondary antibody working well with other primary antibodies? I hope this information is helpful, thank you for your cooperation. Should the suggestions not improve the results, please do not hesitate to contact me again with the further requested details.
