Target

  • Relevance

    There are four IgG subclasses (IgG1, 2, 3 and 4) in humans, named in order of their abundance in serum (IgG1 being the most abundant). IgG2 is the only IgG subclass which passes through the placenta at a level generally lower than that found in the mother. A deficiency of IgG2 indicates a poor antibody response to bacterial polysaccharides and can lead to increased susceptibility to infections caused by encapsulated bacteria.

      Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of human tonsil tissue sections labeling IgG2 with purified ab134050 at 1:40000 dilution (0.025 µg/ml). Heat mediated antigen retrieval was performed using ab93684 (Tris/EDTA buffer, pH 9.0). ImmunoHistoProbe one step HRP Polymer (ready to use) was used as the secondary antibody. Negative control: PBS instead of the primary antibody. Hematoxylin was used as a counterstain.

Cells were fixed with 4% Paraformaldehyde and permeabilised with 90% Methanol. A Goat anti rabbit IgG (Alexa Fluor® 488, ab150077) secondary antibody was used at 1/2000. Isotype control - Rabbit monoclonal IgG (Black). Unlabeled control - Cell without incubation with primary antibody and secondary antibody (Blue).

      Immunocytochemistry/ Immunofluorescence analysis of Raji (human Burkitt's lymphoma cell line) cells labeling IgG2 with purified ab134050 at 1:100 dilution (10 µg/ml). Cells were fixed in 4% Paraformaldehyde and permeabilized with 0.1% tritonX-100. Cells were counterstained with Ab195889 Anti-alpha Tubulin antibody [DM1A] - Microtubule Marker (Alexa Fluor® 594) 1:200 (2.5 µg/ml). Goat anti rabbit IgG (Alexa Fluor® 488, ab150077) was used as the secondary antibody at 1:1000 (2 µg/ml) dilution. DAPI (blue) was used as nuclear counterstain. PBS instead of the primary antibody was used as the secondary antibody only control.

    Can you please let me know if any of the following Anti- human IgG2 primary antibodies cross react with Rhesus IgG2?    I am also interested in purchasing Anti- human IgG3 and IgG4  primary antibodies that recognize Rhesus IgG3 and IgG4, respectively.

    I have reviewed these products and have been in touch with the labs regarding reactivity with Rhesus as requested. We would not recommend any human IgG2 antibody which we know is directed to the hinge region, such as ab1935, against Rhesus as the homology of these is poor. However with this exception, in general there seems to be good homology between Rhesus and Human IgG subtypes.
