Reproducibility is key to advancing scientific discovery and accelerating scientists’ next breakthrough.
Abcam is leading the way with our range of recombinant antibodies, knockout-validated antibodies and knockout cell lines, all of which support improved reproducibility.
We are also planning to innovate the way in which we present recommended applications and species on our product datasheets, so that only applications & species that have been tested in our own labs, our suppliers or by selected trusted collaborators are covered by our Abpromise™ guarantee.
In preparation for this, we have started to update the applications & species that this product is Abpromise guaranteed for.
We are also updating the applications & species that this product has been “predicted to work with,” however this information is not covered by our Abpromise guarantee.
Applications & species from publications and Abreviews that have not been tested in our own labs or in those of our suppliers are not covered by the Abpromise guarantee.

Our Abpromise guarantee covers the use of ab224181 in the following tested applications.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
|Sandwich ELISA||Use at an assay dependent concentration.
Use: 50 ng/well – 200 ng/well (for Capture); 0.05 μg/mL – 0.2 μg/mL (for Detection).
|IHC||Use a concentration of 2 - 5 µg/ml.|
|ELISA||Use at an assay dependent concentration.|
RelevanceIgG3, like IgG1, activates complement and has an affinity for fc receptors; thus IgG1 and IgG3 are the more effective of the IgG subclasses. An IgG3 deficiency is associated with Wiscott-Aldrich disease, systemic lupus erythematosus, juvenile diabetes mellitus, and peridontal infections. IgG anti-viral antibodies are restricted to IgG subclasses 1 and 3, with IgG3 antibodies being the first to appear in response to infection.

A titer ELISA using ab224181. The plate was coated with different amounts of human IgG3. A serial dilution of ab224181 was used as the primary antibody. An alkaline phosphatase conjugated anti-rabbit IgG as the secondary antibody.
ELISA of human immunoglobulins shows ab224181 only reacted to human IgG3. No cross reactivity with Human IgG1, IgG2, IgG4, IgE, IgD, IgA, mouse IgG, rat IgG, or goat IgG. The plate was coated with 50 ng/well of different immunoglobulins. 200 ng/mL, 50 ng/mL, or 10 ng/mL of ab224181 was used as the primary antibody. An alkaline phosphatase conjugated anti-rabbit IgG was used as the secondary antibody.
Sandwich ELISA using ab224181 as the capture antibody (100ng/well), and a biotinylated anti-human light chains ( κ+ λ) antibody as the detection antibody, followed by an HRP conjugated streptavidin.
Sandwich ELISA using ab224181 as the capture antibody (100ng/well), and a biotinylated anti-human light chains ( κ+ λ) antibody as the detection antibody, followed by a HRP conjugated streptavidin.
