Thank you for your call yesterday and for letting us know about the trouble with ab105229.
I've heard back from the lab, and we have not received other reports of the antibody not working, so it's possible that the protocol can be altered to produce better results. The only alterations that we would suggestare to increasethe antigen retrieval and primary antibody incubation times. We also have not tried the antibody with colon and appendix sections, so if you have any tonsil slides we would suggest using them as a positive control. You may also want to try an antibody that you've previously optimized on the colon or appendix tissue, just to make sure the tissue is not too old.
As we discussed, if the staining results do not improve, I would be happy to send a different IGJ antibody free of charge.
