Target

  • Function
    Inhibits NF-kappa-B by complexing with and trapping it in the cytoplasm. However, the unphosphorylated form resynthesized after cell stimulation is able to bind NF-kappa-B allowing its transport to the nucleus and protecting it to further NFKBIA-dependent inactivation. Association with inhibitor kappa B-interacting NKIRAS1 and NKIRAS2 prevent its phosphorylation rendering it more resistant to degradation, explaining its slower degradation.

  • Detection of IkB beta by Immunoprecipitation in whole cell lysate from Huh-7 cells, using ab7547 at 4 µg/mg lysate for IP and ab1162 at a 1/1500 dilution for subsequent Western blot detection.
    Cells expressed recombinant IkB beta tagged with DDDDK. Protein A was used for the Immunoprecipitation step. Primary antibody incubated with matrix for 4 hours at 4°C.

  • ICC/IF image of ab7547 stained HeLa cells. The cells were 100% methanol fixed (5 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab7547, 1/100 dilution) overnight at +4°C. The secondary antibody (green) was ab96899 Dylight 488 goat anti-rabbit IgG (H+L) used at a 1/250 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

Suite à notre conversation téléphonique d'hier, j’ai contacté notre laboratoire a propos de la concentration d’ab7547. Comme l’anticorps est un antisérum (l’anticorps est non purifié), la concentration donnée sur la fiche technique de 90 mg/ml n’est pas la concentration pour l’anticorps uniquement mais pour toutes les protéines dans l’antisérum. La concentration de l’anticorps n’a pas été déterminée mais est estimé à environ 1-10mg/ml.

Thank you for your enquiry and your patience. Anti- IKBbeta antibody may react non-specifically with other proteins. We don't know if you have ordered the control peptide (Ab7548) which will compete only with the specific reaction of antiserum with IKBbeta. We would like to mention that Ab7547 has been reported to cross-react with an undefined 46kDa band which is not IKBbeta. It is possible that you see this band.

Thank you for your enquiry. We do not have any information on optimal Western conditions for this antibody, only that it be used at a dilution of 1/1000. I do not know if it is necessary to incubate the blot with primary overnight. Our Western blot protocol suggests incubating for 1 h at room temperature with agitation; I would recommend trying that first. If you have any more questions, please contact us again.
