Target

  • Function
Produced by activated macrophages, IL-1 stimulates thymocyte proliferation by inducing IL-2 release, B-cell maturation and proliferation, and fibroblast growth factor activity. IL-1 proteins are involved in the inflammatory response, being identified as endogenous pyrogens, and are reported to stimulate the release of prostaglandin and collagenase from synovial cells.
  • Sequence similarities
    Belongs to the IL-1 family.
  • Domain
The similarity among the IL-1 precursors suggests that the amino ends of these proteins serve some as yet undefined function.
  • Cellular localization
Secreted. The lack of a specific hydrophobic segment in the precursor sequence suggests that IL-1 is released by damaged cells or is secreted by a mechanism differing from that used for other secretory proteins.

  • ab7632 staining IL-1 alpha in murine RAW 264.7 cells by Immunocytochemistry/ Immunofluorescence.Cells were fixed in paraformaldehyde, permeabilized using 0.1% Triton-X100 in 2% BSA for 15 minutes, blocked with 2% BSA for 1 hour at 4°C and then incubated with ab7632 at a 1/100 dilution for 16 hours at 4°C. The secondary used was an Alexa-Fluor 488 conjugated chicken anti-rabbit IgG (H+L) used at a 1/500 dilution.

  • Immunohistochemical analysis of pancreatic ductal adenocarcinoma tissue, staining IL-1 alpha with ab7632.

Antigen retrieval was performed by heat mediation in citrate buffer (pH 6.0). Tissue was blocked with 1% BSA for 10 min. The samples incubated overnight with primary antibody (1/40). Staining was detected using DAB. Sections were counterstained with hematoxylin.

Answer

Thank you for contacting us. We are happy to help customers find the most positive control for their research purposes; however we are not specialized in this particular field and do not want to recommend a control that may be unsuitable. I believe that THP-1 cells can be used as positive control since they produce IL1. But I think that they also have to be stimulated in the right way to achieve proper IL1 production. This has not been tested experimentally by us. Maybe this publication will provide more information about this: http://onlinelibrary.wiley.com/doi/10.1002/ddr.1171/abstract We encourage customers to consult the latest literature available through PubMed and other resources in order to find the most up-to-date information about their specific research interests. I am sorry that I could not be more helpful, but I hope that the available literature in this area can provide some clarification. Please do not hesitate to contact us again with other needs or with any questions about our products.

Answer

Thank you for your message.

I am sorry to confirm that this antibody has not been specifically tested on human oral cancer tissue as far as I am aware.

I have done a search and I am sorry I cannot find information to confirm the expression of IL-1 alpha from oral cancer tissue. I would also expect this may depend on the specific type of oral cancer, and I can suggest the customer could consider a further literature search to confirm more details before they start their project.

I can also recommend that if they are not sure of the expression level of IL-1 alpha in their samples, then it would be important to include a positive control tissue sample such as human lymph node or thymus. We have slides available:

ab29871 Lymph node (Human) Tissue Lysate - adult normal tissue
https://www.abcam.com/index.html?datasheet=29871 (or use the following: https://www.abcam.com/index.html?datasheet=29871).

ab30146 Thymus (Human) Tissue Lysate - adult normal tissue
https://www.abcam.com/index.html?datasheet=30146 (or use the following: https://www.abcam.com/index.html?datasheet=30146).

I hope this will be helpful to you. If you have any further questions, please do not hesitate to contact me. I look forward to receiving the completed questionnaire.

Answer

I am not aware that the positive control directions of this antibody have been updated, we currently recommend: Supernatants or lysates of 2 x 106 endotoxin-stimulated human peripheral blood mononuclear cells (PBMC)(PBMC stimulated for 24 hours with 1% (v/v) human serum plus 10 ng/mL E.coli LPS). For use as a positive control. As I mentioned in my previous email the following study used pancreatic tissue from cancerous tissue. IL1-alpha was not present in non-cancerous tissue. As IL-1 alpha is involved in the inflammatory response it may be that it is not abundantly present in normal tissue but I have found reference to both spleen tissue and human fetal skin being used as positive controls. Tjomsland V et al. Interleukin 1a sustains the expression of inflammatory factors in human pancreatic cancer microenvironment by targeting cancer-associated fibroblasts. Neoplasia 13:664-75 (2011).. PubMed: 21847358 The following products may therefore be of use as positive controls: https://www.abcam.com/Pancreas-tumor-human-adenocarcinoma-tissue-slides-ab5036.html https://www.abcam.com/Skin-human-fetal-normal-tissue-slides-ab4417.html and https://www.abcam.com/Spleen-human-normal-tissue-slides-ab4372.html I hope this information has been of help. If you have any further questions please do not hesitate to contact us again.     I hope this information has been of help. If you have any further questions please do not hesitate to ask.

Answer

This antibody has been used for staining of paraffin embedded human pancreas tissue: Tjomsland V et al. Interleukin 1a sustains the expression of inflammatory factors in human pancreatic cancer microenvironment by targeting cancer-associated fibroblasts. Neoplasia 13:664-75 (2011).  PubMed: 21847358 This study used tissue sections from pancreatic ductal adenocarcinoma patients. I am sorry that this has not been reflected on the datasheet. I am in the process of amending this. I am therefore not able to provide a conventional testing discount, however as the datasheet does not currently have any images presented for this application I can offer a testing discount if the customer agrees to provide an image through the Abreview system. If your customer would be interested in this please let me know and I will issue the testing discount.

Question
Answer

Thank you for your call today and for your interest in these antibodies. Since we don't have any TNF alpha, IL1 alpha, or IL10 antibodies tested in squirrel samples, I can offer discounts off a future purchase if you buy these antibodies now, test squirrel, and submit feedback to us in the form of an Abreview. Each discount would be worth 1 free primary antibody. If you are interested in this offer, please follow these steps: 1. Reply to this e-mail to let me know that you would like to proceed and test these antibodies in squirrel. I will then send a discount code for each antibody. These codes must be issued before purchasing the antibodies so please wait for my reply before ordering. 2. Purchase the antibodies either by phone, fax, or online (www.abcam.com). 3. Test in squirrel. 4. Let us know the results, positive or negative, using our Abreview system (this will take about 10 minutes and images are great if you have them!). To find out how to submit an Abreview, please visit: https://www.abcam.com/abreviews. 5. After each review is submitted to us, the discount code becomes active. Simply place your new order by phone, fax, or on the web and mention the discount code. Each discount can be redeemed for any primary antibody ordered and the discount code is valid for 4 months after issue. We are always pleased to obtain feedback about our products and any information is greatly appreciated! Even if an antibody turns out to be unsuitable for squirrel, you will still receive the discount on your next purchase after your Abreview has been submitted. Please let me know if you have any questions about this offer and I would be happy to help you further. The Terms and Conditions of this offer can be found at: www.abcam.com/collaborationdiscount.
