Target

  • Function

    Produced by activated macrophages, IL-1 stimulates thymocyte proliferation by inducing IL-2 release, B-cell maturation and proliferation, and fibroblast growth factor activity. IL-1 proteins are involved in the inflammatory response, being identified as endogenous pyrogens, and are reported to stimulate the release of prostaglandin and collagenase from synovial cells.

    The similarity among the IL-1 precursors suggests that the amino ends of these proteins serve some as yet undefined function.

    Thank you for contacting us. We are happy to help customers find the most positive control for their research purposes; however we are not specialized in this particular field and do not want to recommend a control that may be unsuitable. I believe that THP-1 cells can be used as positive control since they produce IL1. But I think that they also have to be stimulated in the right way to achieve proper IL1 production. This has not been tested experimentally by us. Maybe this publication will provide more information about this: http://onlinelibrary.wiley.com/doi/10.1002/ddr.1171/abstract We encourage customers to consult the latest literature available through PubMed and other resources in order to find the most up-to-date information about their specific research interests.     I am sorry to confirm that this antibody has not been specifically tested on human oral cancer tissue as far as I am aware.

    I have done a search and I am sorry I cannot find information to confirm the expression of IL-1 alpha from oral cancer tissue. I would also expect this may depend on the specific type of oral cancer, and I can suggest the customer could consider a further literature search to confirm more details before they start their project.

    I can also recommend that if they are not sure of the expression level of IL-1 alpha in their samples, then it would be important to include a positive control tissue sample such as human lymph node or thymus. We have slides available:

    I am not aware that the positive control directions of this antibody have been updated, we currently recommend: Supernatants or lysates of 2 x 106 endotoxin-stimulated human peripheral blood mononuclear cells (PBMC)(PBMC stimulated for 24 hours with 1% (v/v) human serum plus 10 ng/mL E.coli LPS). For use as a positive control. As I mentioned in my previous email the following study used pancreatic tissue from cancerous tissue. IL1-alpha was not present in non-cancerous tissue. As IL-1 alpha is involved in the inflammatory response it may be that it is not abundantly present in normal tissue but I have found reference to both spleen tissue and human fetal skin being used as positive controls. Tjomsland V et al. Interleukin 1a sustains the expression of inflammatory factors in human pancreatic cancer microenvironment by targeting cancer-associated fibroblasts. Neoplasia 13:664-75 (2011)..     This antibody has been used for staining of paraffin embedded human pancreas tissue: Tjomsland V et al. Interleukin 1a sustains the expression of inflammatory factors in human pancreatic cancer microenvironment by targeting cancer-associated fibroblasts. Neoplasia 13:664-75 (2011).  PubMed: 21847358 This study used tissue sections from pancreatic ductal adenocarcinoma patients. I am sorry that this has not been reflected on the datasheet. I am in the process of amending this. I am therefore not able to provide a conventional testing discount, however as the datasheet does not currently have any images presented for this application I can offer a testing discount if the customer agrees to provide an image through the Abreview system.     Thank you for the information on this antibody discount program.  We would like to start with one antibody; TNF alpha, and see how it goes.  TNF alpha has the best homology between squirrel and human of the initial three targets we are interested in.  We would like to try Ab6671 which is an anti-TNF alpha rabbit polyclonal with recombinant full length human protein as the immunogen. As it happens, two of the post-docs in our lab have recently purchased antibodies from Abcam which they have used in our squirrels and are happy to write an abreview for each of these.  Is it possible for them to receive credit for future purchases based on these potential reviews?  The two antibodies are beta-catinen (Ab16051) and BDNF (Ab72439) and both were used for Western Blots.
