IHC-P 1/200. (see Abreview)
IP Use at an assay dependent concentration. This antibody has been used to immunoprecipitate >90% human IL-1 (in solution using a double-antibody method).
ELISA Use a concentration of 2 µg/ml. This antibody has been used as a solution phase antibody in a two site ELISA with biotin conjugated IL-1 alpha antibody. Typically the ELISA has a working range of 0-10000 pg/ml and a sensitivity of 5 pg/ml with the appropriate standard. Recommended sample volumes are 25-200 µl. Recovery of human IL-1 alpha in human serum is typically >85% in the ELISA.
Neutralising Use a concentration of 25 mg/ml. This antibody has been used to neutralize >95% of the biological activity of human IL-1 at 0.5 mg/ml.
IHC (PFA fixed) Use a concentration of 1 - 10 µg/ml. This antibody has been used to stain human IL-1 alpha expressed in mononuclear cells, following fixation with 4% paraform in PBS for 10 minutes at 4oC plus 0.1% saponin 20 minutes at RT.


  • Function
    Produced by activated macrophages, IL-1 stimulates thymocyte proliferation by inducing IL-2 release, B-cell maturation and proliferation, and fibroblast growth factor activity. IL-1 proteins are involved in the inflammatory response, being identified as endogenous pyrogens, and are reported to stimulate the release of prostaglandin and collagenase from synovial cells.

    Belongs to the IL-1 family.
  • Domain
    The similarity among the IL-1 precursors suggests that the amino ends of these proteins serve some as yet undefined function.
  • Cellular localization
    Secreted. The lack of a specific hydrophobic segment in the precursor sequence suggests that IL-1 is released by damaged cells or is secreted by a mechanism differing from that used for other secretory proteins.

  • ab17281 staining IL1 alpah in human colon tissue by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections). Tissue was fixed with paraformaldehyde and a heat mediated antigen retrieval step was performed using EDTA buffer pH 9.0. Samples were then blocked with 10% serum for 30 minutes at 21°C followed by incubation with the primary antibody at a 1/200 dilution for 30 minutes at 21°C. A undiluted HRP-conjugated goat polyclonal was used as secondary antibody.

The details you have kindly provided will enable us to investigate this case for you and this is also helpful in our records for monitoring of quality. Reviewing this case, I would like to offer some suggestions to help optimise the results from ab17281. I would also appreciate if you can confirm some further details: 1. For antigen retreival, I can suggest to continue to try heat mediated retreival with EDTA pH9, as this has been used succesfully in an Abreview submitted by another customer (published on the datasheet). However, I woudl recommend to try different time points as this may help to optmize the results. Try heating for 1, 3,5, 10 and 20 minutes. 2. I can recommend to try incubating in PBS containing 0.2% Tween. The Tween will help to solubilise the antibody. It will also help to permeablize the cells to allow staining of any IL1 alpha in the cytoplasm. 3. PBS continaing 0.2% tween can also be used as a wash buffer. Wash 3 times only at each appropriate wash step. 4. Could you confirm if the current vial of secondary antibody has been used succesfully with other primary antibodies? In the event that a product is not functioning in the applications cited on the product data sheet (and the problem has been reported within 6 months of purchase), we will be pleased to provide a credit note, free of charge replacement or refund.
