  • Function
    Potent proinflammatory cytokine. Initially discovered as the major endogenous pyrogen, induces prostaglandin synthesis, neutrophil influx and activation, T-cell activation and cytokine production, B-cell activation and antibody production, and fibroblast proliferation and collagen production. Promotes Th17 differentiation of T-cells.

    Activation of the IL1B precursor involves a CASP1-catalyzed proteolytic cleavage. Processing and secretion are temporarily associated.

In response to inflammasome-activating signals, such as ATP for NLRP3 inflammasome or bacterial flagellin for NLRC4 inflammasome, cleaved and secreted. IL1B lacks any known signal sequence and the pathway(s) of its secretion is(are) not yet fully understood (PubMed:24201029). On the basis of experimental results, several unconventional secretion mechanisms have been proposed. 1. Secretion via secretory lysosomes: a fraction of CASP1 and IL1B precursor may be incorporated, by a yet undefined mechanism, into secretory lysosomes that undergo Ca(2+)-dependent exocytosis with release of mature IL1B (PubMed:15192144). 2. Secretory autophagy: IL1B-containing autophagosomes may fuse with endosomes or multivesicular bodies (MVBs) and then merge with the plasma membrane releasing soluble IL1B or IL1B-containing exosomes (PubMed:24201029). However, autophagy impacts IL1B production at several levels and its role in secretion is still controversial. 3. Secretion via exosomes: ATP-activation of P2RX7 leads to the formation of MVBs containing exosomes with entrapped IL1B, CASP1 and other inflammasome components. These MVBs undergo exocytosis with the release of exosomes. The release of soluble IL1B occurs after the lysis of exosome membranes (By similarity). 4. Secretion by microvesicle shedding: activation of the ATP receptor P2RX7 may induce an immediate shedding of membrane-derived microvesicles containing IL1B and possibly inflammasome components. The cytokine is then released in the extracellular compartment after microvesicle lysis (PubMed:11728343). 5. Release by translocation through permeabilized plasma membrane. This may occur in cells undergoing pyroptosis due to sustained activation of the inflammasome (By similarity). These mechanisms may not be not mutually exclusive.

  • Immunohistochemical analysis of frozen Human skin tissue taken from patients with localized cutaneous leishmaniasis (A, C), diffuse cutaneous leishmaniasis (E). Normal tissue (G) was used as a negative control.

    IL-1 beta was stained using ab8320 at 1/100 dilution for 1 hour at room temperature. ab94705 Mouse and Rabbit Specific HRP/AEC Detection IHC Kit was used to detect staining. Hematoxylin was used as a counterstain.

  • ab8320 staining IL-1 beta in lung inflammatory cells from Rhesus monkey by ICC/IF (Immunocytochemistry/immunofluorescence). Cells were fixed with formaldehyde, permeabilized with Tween 20 and blocked with 10% serum for 2 hours at 21°C. Samples were incubated with primary antibody (1/25 in PBS + 10% serum + 1% Tween 20) for 18 hour at 4°C. An diluted Alexa Fluor®594-conjugated + 1.4nm Goat anti-mouse IgG polyclonal was used as the secondary antibody.

I am glad that the Rhesus tissue worked well! Unfortunately the homology for ab8320 with sheep is only 60%, to low for any reactivity. Another IL1-beta antibody, ab45692 does have 88% homology but has only been tested for Western Blot. Please let me know if I can be of further assistance.

Thank you very much for contacting us. To our knowledge, ab8320 has not been tested in rhesus monkey. The immunogen for this product is full length human IL1 beta, which has more than 90% homology with the Rhesus sequence. Based on this, I would predict that it would react, but of course cannot guarantee it.

Para detectar IL1 beta por Sándwich ELISA, ab8320 es una buena opción, como puedes ver en su ficha técnica, es una aplicación testada. Pero en aquellos anticuerpos testados en sELISA, en la datasheet siempre se indicará si dicho anticuerpo actúa como captura o detección, y asimismo se indicará el anticuerpo par con el que se ha testado. En el caso de ab8320, éste se ha testado en sELISA conjuntamente con ab8321, conjugado a la enzima HRP.

Si lo que desea el cliente es hacer un ensayo de ELISA indirecto, ab8320 también puede usarse, y el secundario propuesto, al haberse usado también en ELISA, es una buena opción.

Sin embargo, la proteína sugerida no es la más adecuada para trabajar con estos anticuerpos. Como verás en su datasheet, en el apartado de Application Notes, otros primarios se sugieren para realizar un ensayo ELISA: ab18642 y ab18637.

Por tanto para el caso de IL1B yo recomendaría la proteína ab9617 con uno de los anticuerpos sugeridos, y el secundario también indicado.
