  • Function

    Cytokine that stimulates the proliferation of T-lymphocytes. Stimulation by IL-15 requires interaction of IL-15 with components of IL-2R, including IL-2R beta and probably IL-2R gamma but not IL-2R alpha.

    Most abundant in placenta and skeletal muscle. It is also detected in the heart, lung, liver and kidney. IL15-S21AA is preferentially expressed in tissues such as testis and thymus.

As discussed over the phone, I have been looking into how the anti-IL15 antibodies ab40668 and ab134177 have been tested and whether they would be the most suitable antibodies to use in your experiments.

As discussed over the phone, the bands observed with ab40668 and ab134177 in Western blotting are not exactly where they would be expected to run as recombinant fusion proteins were used. With ab40668 as fusion protein of IL-15 (aa49-162, 12kDa) and Thioredoxin-1 (aa1-109, 12kDa), plus His tag (7kDa) was used, bringing the expected molecular weight to ˜ 31 kDa which is what is observed on the blot. The antibody ab134177 used a slightly truncated version of the IL15, making the expected band size ˜12 kDa (as observed).

Unfortunately, both of these antibodies have been tried with samples containing endogenously expressed IL15 and we have been unable to detect a signal. It may be that the conditions necessary for using these antibodies with endogenous samples requires some further optimisation. I would suggest, as you are intending to use the antibodies with endogenous samples to have a look at the ab7213 or ab109082 instead. Both of these would be covered by the Abpromise guarantee for the application and species you intend to use the antibody for.
