  • Immunohistochemical analysis of formaldehyde-fixed paraffin-embedded murine obsese nephric tissue sections, labelling IL-18 with ab71495 at a dilution of 1/2500 incubated for 12 hours at 4°C in PBS and 2.5% horse serum diluent. Heat mediated antigen retrival was with citrate buffer and blocking was with 2.5% horse serum in PBS incubated for 1 hour at 25°C. Secondary was a horse anti-rabbit polyclonal HRP conjugate.

  • IHC image of ab71495 staining in mouse normal spleen formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab71495, 1µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

    For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

I am sorry to confirm that as far as we are aware, ab71495 has never been tested with samples from dog. All tested species cross-reactivity information covered by our guarantee is stated on our datasheets, and these are updated as soon as any new information is brought to our attention.

I can confirm the immunogen is from the mouse protein, taken from between amino acids 25 -65 (this antibody issourced externally and regrettably in this case the exact sequence is proprietary). This sequence has 60% alignment to the dog sequence. We recommend that alignment should be 85% to predict that an antibody will detect in a different species. In this case, the alignment is low, so althoughthere is a possibility of some weak crossreactivity, unfortunately I would not suggest this antibody is likely to work well in dog.

Unfortunately, the exact peptide sequence information is also not available (proprietary and can not be released). However, the originators of the product were willing to release the amino acid sequence range from where the exact peptide sequence was taken. The immunogen is a sequence of amino acids between 1- 157 of mature mouse. I hope this information will still be helpful.
