  • Application notes
    ELISA: Use at a concentration of 1 µg/ml to detect 0.6 ng/well of recombinant human IL2.
    Neut: Use at a concentration of 0.8 - 8.0 µg/ml.
    WB: Use at a concentration of 2 µg/ml, this detects 5 ng/lane of recombinant human IL2 under reducing conditions and 20 ng/lane under non-reducing conditions.Predicted molecular weight: 19 kDa.

    Anti-Human IL2 is tested for its ability to neutralize the biological activity of recombinant, human IL2 on CTLL2 cells. The ND50 of the antibody is defined as the concentration of antibody resulting in a one-half maximal inhibition of bioactivity of recombinant, human IL2, when IL2 is present at a concentration just high enough to elicit a maximum response.

      Produced by T-cells in response to antigenic or mitogenic stimulation, this protein is required for T-cell proliferation and other activities crucial to regulation of the immune response. Can stimulate B-cells, monocytes, lymphokine-activated killer cells, natural killer cells, and glioma cells.

      Note=A chromosomal aberration involving IL2 is found in a form of T-cell acute lymphoblastic leukemia (T-ALL). Translocation t(4;16)(q26;p13) with involves TNFRSF17.

    Thank you for forwarding some details of your protocol following our conversation this morning. I understand from the questionnaire you kindly sent that you have only attempted a sandwich ELISA experiment with ab10752. This application has not been tested, therefore it is very hard to know what the problem might be, and I am unable to guarantee that the antibody works in sandwich ELISA. Have you tried a direct ELISA as described on the datasheet? I am also concerned that the levels of IL2 in your samples are low, if you have not done so already I would recommend to run a positive control. You do not mention many details of your experiments in regards to the antibody (dilution, buffer..etc), blocking and times, and I would be more than happy to provide protocol suggestions, but from what I understand you do not have any antibody left and do not wish to continue using this product. I'm sorry that I cannot offer you a replacement vial or refund in this instance, as we discussed on the phone this is because we only guarantee our products on applications tested.
