    Genetic variations in IL2RA are associated with susceptibility to diabetes mellitus insulin-dependent type 10 (IDDM10) [MIM:601942]. A multifactorial disorder of glucose homeostasis that is characterized by susceptibility to ketoacidosis in the absence of insulin therapy. Clinical fetaures are polydipsia, polyphagia and polyuria which result from hyperglycemia-induced osmotic diuresis and secondary thirst. These derangements result in long-term complications that affect the eyes, kidneys, nerves, and blood vessels.

The lymphocytes are obtained from blood sample using Ficoll Hypaque histopaque 1077 (reference from Sigma H8889, but available from other suppliers) according with the protocol given by the supplier.
The PHA is a leucoagglutinin (reference from Sigma L4144, but available from other suppliers) and we use it around 1 µg/ml/10E6 cells during 72h. The concentration given is a medium use, each batch needs to be validated, the concentration optimal used is different one batch to another (try a range around the value given).
The activation could be done in 96 wells or in flask also, depending of the cell quantities needed and the experiments done after the test.
