Target

  • Function
    Receptor for interleukin-2.
  • Involvement in disease
Genetic variations in IL2RA are associated with susceptibility to diabetes mellitus insulin-dependent type 10 (IDDM10) [MIM:601942]. A multifactorial disorder of glucose homeostasis that is characterized by susceptibility to ketoacidosis in the absence of insulin therapy. Clinical fetaures are polydipsia, polyphagia and polyuria which result from hyperglycemia-induced osmotic diuresis and secondary thirst. These derangements result in long-term complications that affect the eyes, kidneys, nerves, and blood vessels.

  • Overlay histogram showing Jurkat cells stained with ab8235 (red line). The cells were fixed with 80% methanol (5 min) and incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab8235, 1µg/1x106 cells) for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (ab91353, 2µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed. This antibody gave a positive result in 4% paraformaldehyde (10 min) fixed Jurkat cells used under the same conditions.
Please note that Abcam do not have any data for use of this antibody on non-fixed cells. We welcome any customer feedback.

Answer

The antibody ab8232 is unconjugated and so you would need to use a secondary antibody for detection. If you tell me what techniques you are using I would be happy to make some suggestions about suitable secondary antibodies. Please let me know if there is anything else I can help you with.

Answer

Thank you for your enquiry. I ran a search for a neutralising antibody directed against the IL2 Receptor alpha and unfortunately could find any results. This does not mean that the ab8235 will not work in blocking IL2, it just means that we have not tested any of the IL2 Receptor alpha in neutralisation. If you do decide to purchase the antibody for blocking, we can not guarantee that it will work, but please let us know how it goes! You can earn abpoints towards many promotions by filling out Abreviews about your experience. Also, at the bottom of any search results page, there is a link to the World Antibody Gateway which can search 249 online antibody companies. I hope this information helps, please do not hesitate to contact us if you need any more advice or information.

Answer

Thank you for your message. We are very sorry but there was a discrepancy on our datasheet. We would like to confirm that this antibody has not been tested and characterised for Western blot application. Thank you for your feedback; it is very likely that the antibody recognizes only the native form of the protein and it is not suitable for WB. We can offer you either a credit note or a refund. Would you be so kind to let us know how you would prefer to proceed? Please could you confirm your Purchase Order Number or your Abcam Order Number. We apologize for any inconvenience caused and thank you for bringing this matter to our attention.

Answer

Our datasheet currently states that the antibody has not yet been tested in WB and I apologise if it previously did; I have checked with our source and they confirmed they have not tested it yet. Should you wish a refund can you please give me your order number and/or Abcam order number and I will arrange this straight away. If you still wish to test ab8235 I would recommend a preliminary experiment to test which dilution would be best: try 1:200, 1:500, 1:1000 and 1:2000 and optimise the dilution according to your results. Please let us know how you get on with ab8235 in WB and in return we will forward a GBP10/ USD15/ EUR15 Amazon gift voucher. Please do not hesitate to contact me if you need further advice,
