      Genetic variations in IL2RA are associated with susceptibility to diabetes mellitus insulin-dependent type 10 (IDDM10) [MIM:601942]. A multifactorial disorder of glucose homeostasis that is characterized by susceptibility to ketoacidosis in the absence of insulin therapy. Clinical fetaures are polydipsia, polyphagia and polyuria which result from hyperglycemia-induced osmotic diuresis and secondary thirst. These derangements result in long-term complications that affect the eyes, kidneys, nerves, and blood vessels.

        Flow Cytometry - Anti-IL-2 Receptor alpha antibody [MEM-181] (ab8235)
        Overlay histogram showing Jurkat cells stained with ab8235 (red line). The cells were fixed with 80% methanol (5 min) and incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab8235, 1µg/1x106 cells) for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (ab91353, 2µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed. This antibody gave a positive result in 4% paraformaldehyde (10 min) fixed Jurkat cells used under the same conditions.
        Please note that Abcam do not have any data for use of this antibody on non-fixed cells. We welcome any customer feedback.

      Question

      I am working on a IL2R construct. I will transfect cells, and then use the antibody ab8235 for immunocytochemistry. the IL2R protein is supposed to be localized to the plasma membrane. I went over abreview of this product, and I realized that the lab that has used it for ICC, had permealized the cells with 0.5% Triton X-100 or 0.5% Saponin. My question is do I really need the permeabilisation. As the IL2R chimeric construct is supposed to be present on the cell membrane, then I would expect the antibody to bind to the protein without permeabilisation!! I actually need to to skip the permeabilisation step, because I want to try the used the anitbody for construct clustering. another question that might help me get an answer, is whether the epitope is extracellular or not. thanks

      Thank you for your enquiry. I ran a search for a neutralising antibody directed against the IL2 Receptor alpha and unfortunately could find any results. This does not mean that the ab8235 will not work in blocking IL2, it just means that we have not tested any of the IL2 Receptor alpha in neutralisation. If you do decide to purchase the antibody for blocking, we can not guarantee that it will work, but please let us know how it goes!       We have used the antibody but have not been successful in Western blotting. It is likely that the antibody is conformation senstive and the rigours of SDS and mercaptoathanol have rendered it non-recognisable.

      Our datasheet currently states that the antibody has not yet been tested in WB and I apologise if it previously did; I have checked with our source and they confirmed they have not tested it yet.
