1.) Ich kann empfehlen eine Fixierung mit PFA 4% für 10 Minuten und eine Fixierung mit Methanol noch auszuprobieren. In der Tat haben verschieden Fixierungsmethoden oft einen grossen Einfluss auf die Immunomarkierung. Zum Beispiel fixiert PFA durch kovalentes Krosslinken von Proteinen und Methanol/Aceton durch Präzipitieren von den Proteinen. Dies kann die Epitope unterschiedlich maskieren/demaskieren und daher sollte es für jeden Antikörper optimisiert werden.
2.) Ich kann auch empfehlen doch eine Verdünnung von 1/100 mal auszuprobieren, da die Sensibilität vielleicht tiefer ist bei gefrorenen Schnitten.

I am sorry to see that your customer is having so much difficulty with so many antibodies. Can you please ask your customer to clarify the fixation step? They listed "xylol" as the fixative, which I expect is just a misunderstanding. Xylol is typically used to remove paraffin, not to fix tissues. Most labs use a formaldehyde fixative, such as a solution of PFA or formalin. Are the samples all sections of paraffin-embedded tissue? The one suggestion I have is for the customer to test different amounts of time for the antigen retrieval step. Two minutes may not be long enough or for some antibodies, not retrieval is necessary and may even be harmful. I recommend testing ab18499 on three tonsil sections that have had either no AR, 2 minutes, or 15 minutes with citrate 10mM, pH6. For other antibodies, the customer may simply need to apply more antibody. For instance, on the datasheet for anti-IL17 ab79056, for the stain of IL17 in dog lymph node, the antibody was used at a dilution of 1/100. Your customer applied it at 1/400 to the tonsil section. Depending on the expression of IL17 in tonsil, the 1/400 dilution may be insufficient. Could you ask the customer to please send you an image of the good IL6 stain, and an image of one of the non-specific stains, such as the ab34843 on tonsil? When you send the customer's reply regarding the xylol, could you attach those images too? If the customer does not have them, that's OK.
