    Associates with IL12B to form the IL-23 interleukin, an heterodimeric cytokine which functions in innate and adaptive immunity. IL-23 may constitute with IL-17 an acute response to infection in peripheral tissues. IL-23 binds to an heterodimeric receptor complex composed of IL12RB1 and IL23R, activates the Jak-Stat signaling cascade, stimulates memory rather than naive T-cells and promotes production of proinflammatory cytokines. IL-23 induces autoimmune inflammation and thus may be responsible for autoimmune inflammatory diseases and may be important for tumorigenesis.

    Human monocytes were fixed in methanol, permeabilized using 0.1% Saponin/ PBS, blocked with 4% serum for 30 minutes at 25°C and then incubated with ab84471 at 5µg/ml for 48 hours at 37°C.

    Staining: IL-17 (red) GFP (green).

    We have examined the effect of IL-23 and in combination with anti-IL-23 on IL-17 production in monocytes. IL-23 leads to increased IL-17 secretion (Figure B and C) and this effect was abolished with anti-IL-23 (Figure D). IL-2 is part of our culture medium for monocytes. IL-2 stimulation serves as a basis to keep the cell alive. Thus almost every cytokine that we used is used in combination with IL-2. The lab has provide me with the neutralization protocol used by them. The peptides and reagents are all listed on this. The test is done on murine cells for human cytokine, the mIL-23 and hIL-23 are closed and cross-reacted on cells from mice or human. I have attached this protocol for your review.

The bovine protein is 81% similar to human protein so the possibility of cross reactivity is high. Unfortunately for monoclonal antibodies we do not know the exact epitope sequence so I am sorry I can't be more specific; I can however still provide you testing discount code.

Thank you for contacting us. I have attached the protocol that we received from the laboratory that tested this IL23 antibody for neutralization. The assay measures the amount of IL17 that is produced by mouse splenocytes in response to IL23 and IL2 protein, +/- the anti-IL23 antibody. (Mouse primary splenocytes were used because results with continuous human cell lines were inconclusive). The measurement of IL17 requires an ELISA. The kit from R&D is suggested but we also have a similar kit, a sandwich ELISA for mouse IL17, with the catalogue number ab100702. For IL23 protein, we have ab106889, and for IL2, ab9856.
