As discussed over the phone I have now looked into which antibodies we have against IL17E which would be likely to cross react with mouse samples. All the antibodies we have against this target are raised against immunogens taken from the human protein.

However, we have a couple of antibodies where the immunogen used to raise the antibody shares high homology with the mouse protein and would therefore be predicted to react. As discussed over the phone, the immunogen used to raise the antibody ab108530 shares only 71% homology and we would therefore not recommend trying this. I have removed the reference to "predicted to react" with mouse for this antibody to avoid future confusion.

I have found that the mouse monoclonal antibody ab13651 has actually already been tested with mouse testis samples and found to detect a band between 14-21 kDa in Western blotting. I would therefore be able to offer the Abpromise for this antibody. This would mean that if you purchased this antibody and you were not able to get good results with mouse samples you would be eligible for a replacement antibody of your choice, a credit note or refund.

I understand you would prefer an antibody raised in rabbit and we do have one that may be worth trying. The rabbit polyclonal ab123666 has been raised against the amino acid residues 74-104 of human IL17E which matches exactly to the mouse protein (SwissProt reference Q8VHH8) apart from 3 residues. We would therefore expect the antibody to be able to cross react. If you would like to participate in the testing discount scheme with this antibody I can raise the discount code for you.

For ab17613, my colleague has confirmed that it is sterile filtered and lyophilized from PBS, pH 7.2 and is recommended to be reconstituted with sterile water to a concentration of 0.1-1.0mg/mL. Regarding the unsuccessful conjugation, she recommended checking the pH of the reconstituted antibody to ensure that it is within the tolerance of the kit. She also suggested optimizing the amount of dye to ensure the appropriate molar ratio.
