The tissue was acetone fixed and embedded into GMA resin.
The primary antibody is incubated overnight at room temperature.
We currently use another IL-8 antibody, clone [NAP II], which has recently been discontinued. This was used alongside the Abcam IL-8 as a positive control and it gave good strong positive staining.
The Abcam IL-8 antibody was tried at serial dilutions from 1:10 to 1:160, with no positive staining. It was tried a second time on different pieces of nasal polyp and tonsil at dilutions of 1:10 and 1:100 and was still negative.
