  • Indoleamine 2 3-dioxygenase antibody (ab55305) at 1ug/lane + HeLa cell lysate at 25ug/lane.

This image was generated using the ascites version of the product.

  • Overlay histogram showing HeLa cells stained with ab55305 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab55305, 1µg/1x106 cells) for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was mouse IgG2b [PLPV219] (ab91366, 2µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed.

This image was generated using the ascites version of the product.

Thank you for contacting us.

I have found a research paper about Indoleamine 2,3-Dioxygenase tissue distribution and cellular localization that may be useful to determine the cellular localization.

Indoleamine 2,3-Dioxygenase Tissue Distribution and Cellular Localization in Mice: Implications for Its Biological Functions
Xiangchen Dai, Bao Ting Zhu

We have online protocols for both IHC-P and IHC-Fr . I would suggest testing the standard protocol and optimizing if required.

If performing IHC-P, a concentration of 3ug/ml is advised, although this may need optimization. We also have a paper referencing this product in IHC-Fr.

Blunted epidermal L-tryptophan metabolism in vitiligo affects immune response and ROS scavenging by Fenton chemistry, part 2: Epidermal H2O2/ONOO(-)-mediated stress in vitiligo hampers indoleamine 2,3-dioxygenase and aryl hydrocarbon receptor-mediated immune response signaling.

Thank you for your email.

I had sent a reply to you 5 minutes before I received your second email. Our emails must have crossed each other on the way.

For ab134197, please see my previous email.

For ab106134, this antibody would be guaranteed for mouse and rat samples as well as IHC-P. The complaint you saw, could be from the same customer. Nevertheless, the antibody would be guaranteed. Sometimes a vial or a lot might not be working for a customer and the cause is not always easy to decipher.

As for ab27924 and ab55305, they both have been tested with human samples in IHC-P. Mouse and rat samples have not yet been tested. Thus, you could use our testing discount program (see my previous email for the details) for trying the antibodies in mouse and rat.

I hope this information is helpful to you. Please do not hesitate to contact us if you need any more advice or information.
