  • Overlay histogram showing HeLa cells stained with ab55305 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab55305, 1µg/1x106 cells) for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was mouse IgG2b [PLPV219] (ab91366, 2µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed.

We have online protocols for both IHC-P and IHC-Fr . I would suggest testing the standard protocol and optimizing if required.

If performing IHC-P, a concentration of 3ug/ml is advised, although this may need optimization. We also have a paper referencing this product in IHC-Fr.

For ab134197, please see my previous email.

For ab106134, this antibody would be guaranteed for mouse and rat samples as well as IHC-P. The complaint you saw, could be from the same customer. Nevertheless, the antibody would be guaranteed. Sometimes a vial or a lot might not be working for a customer and the cause is not always easy to decipher.

As for ab27924 and ab55305, they both have been tested with human samples in IHC-P. Mouse and rat samples have not yet been tested. Thus, you could use our testing discount program (see my previous email for the details) for trying the antibodies in mouse and rat.
