Target

  • Relevance

    The nucleoprotein (NP) of Influenza virus encapsulates the negative strand of the viral RNA and is essential for replicative transcription. It may also be involved in other essential functions throughout the virus life cycle. As well as binding ssRNA, NP is able to self associate to form large oligomeric complexes. NP is able to interact with a variety of other macromolecules of both viral and cellular origins. It binds the PB1 and PB2 subunits of the polymerase and the matrix protein M1. "NP has also been shown to interact with at least four cellular polypeptide families: nuclear import receptors of the importin class, filamentous (F) actin, the nuclear export receptor CRM1 and a DEAD box helicase BAT1/UAP56" (Portela et al 2002).

      Immunofluorescence assay of MDCK cells derived from canine kidney cells, and A549 cells derived from human lung carcinoma cells, that were infected with H1N1 influenza virus (A/PuertoRico/8/34). Samples were taken at 3, 9, and 24 hours post-infection. Influenza A Virus Nucleoprotein antibody (ab128193) efficiently detected virus-infected MDCK and A549 cells as early as 3 h after infection. The cells were fixed with 4% paraformaldehyde in phosphate-buffered saline (PBS) and permeabilized with 0.1% 0.1% Triton X-100 in PBS. The bound antibody was visualized by a further reaction with an green flurescent secondary antibody.

    Please see below response from customer on your additional queries-

    I am surprised to know the information provided by us still not sufficient to troubleshoot.
    1. By the way the chemicon antibody is only for IFA applications. If chemicon has antibody with western blot and immunoprecipitation applications, perhaps I wouldn't be getting abcam antibody.

    2. the construct contains full NP sequence without any tag.

    3. We use H1N1, H3 N2 strains, in addition I have several seasonal isolated. all the strains been confirmed. But for this assay we used H1N1 and H3N2 strains.

    4. For your kind information in my lab every student does western blot with same detection system. We do not have any problem with detection method.

    5. I do not have purified NP protein. If I have purified protein probably I would have tried to raise antibody in the lab itself.

    Hope this information is sufficient.

    When experiments are carried out using transfected cells, it is very important to provide information about the construct and any positive controls used to make sure that the experiments are nicely controlled. Without these details and the exact strains of the virus, it is hard to troubleshoot and to provide sufficient technical support.

    We have not received any similar complaint about ab128193, thus not aware of any QC issues. Currently, we have the same batch in stock your customer has tried. I could offer you a new vial of ab128193 or one vial of any alternative antibodies against the same target protein (Influenza A Virus Nucleoprotein).

    Dear All,
    I ordered the Anti-Influenza A Virus Nucleoprotein antibody [C43] (ab128193) as you stated (https://www.abcam.com/Influenza-A-Virus-Nucleoprotein-antibody-C43-ab128193.html) that it would have reacted with NP of all influenza A subtypes and the Mouse monoclonal [B247M] to influenza A virus Nucleoprotein (ab66191). I have been testing both of them in Western Blot analysis and none of them worked with the H7N3 avian influenza subtype that I am currently using for my studies. Therefore I had to use another Mab (AbCam) from a different lab. I would ask you to give me you best quotation for this Mab: “ Mouse monoclonal [9G8] to influenza A virus Nucleoprotein (ab43821)”.

    I am sorry if ab128193 did not work as you expected.

    H7N3 subtype of avian Influenza A virus has not been tested with this antibody. I would recommend you to test IF with the virus-infected cells or ELISA with inactivated purified antigen of N7H3 to examine the reactivity of C43 antibody with N7H3 subtype.

    In regards to ab43821, it should recognize H7N3 influenza; however, it was not tested on this viral strain.
