Images

  • ICC/IF image of ab15459 stained HeLa cells (ab150035ab15459), 1µg/ml) overnight at +4°C. The secondary antibody (green) was a goat anti-rabbit Alexa Fluor® 488 (IgG; H+L) ab150077) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

  • Human small intestine (ab29276) stained with anti-Integrin beta 5 antibody, ab15459 by Immunohistochemistry (FFPE-sections).

  • ab15459 staining Integrin beta 5 in Rat gastric epithelial cells by ICC/IF (Immunocytochemistry/immunofluorescence). Cells were fixed with paraformaldehyde, permeabilized with 0.1% Triton and blocked with 0.25% serum free protein for 20 minutes at 28°C. Samples were incubated with primary antibody (1/50 in antibody diluent) for 2 hours at 28°C. An FITC-conjugated Goat anti-IgG polyclonal (1/80) was used as the secondary antibody. Nuclei were counterstained with propidium iodide (ab14083).

Unfortunately, this antibody is not against the extracellular domain and so will not be suitable for your application.

We have not tested any of our antibodies in vivo and therefore cannot guarantee that any of these will work on live unpermeabilised cells.

In addition, I have read through the literature and have been unable to identify other tools which are used for labelling integrin b5 in vivo.

I would like to recommend checking the Biocompare website which has an excellent antibody search facility that includes many suppliers. The links are:

Yes, the blocking buffer that was used to generate that image was a combination of 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilize the cells and block non-specific protein-protein interactions. The permeabilization and blocking steps were done at the same time this way. You can perform this step at room temperature. This is the protocol we've used in house.

We have heard that a customer tried separating out the steps and it worked:


Permeabilization:
Yes - 0.1 % Triton X-100

Blocking step:
BSA as blocking agent for 30 mins · Concentration: 2 % · Temperature: 25°C
