Thank you for your reply. I am very glad to be of help with this. You understand the program perfectly. I have created a code for testing Ab24324 in WB for you below. There is a 4 month window for our 100% Abreview Testing Program within which you must submit the Abreview for ab24324 and redeem the code. I am very pleased to hear you would like to accept our offer and test ab24324 in Western Blot. This code will give you: 1 free PRIMARY ANTIBODY before the expiration date. To redeem this offer, please submit an Abreview for Western Blot and include this code in the “Additional Comments” section so we know the Abreview is for this promotion. Please remember that submission of the Abreview is sufficient for the discount code to become active. For more information on how to submit an Abreview, please visit the site: www.abcam.com/Abreviews. Remember, we publish both positive and negative Abreviews on our datasheets so please submit the results of your tests. The code will be active once the Abreview has been submitted and can be redeemed in one of the following ways: 1) Call to place your order and mention the code to our customer service department; 2) Include the code in your fax order; 3) Place your order on the web and enter the promotional code. Answer

Thank you for your reply. This clone was first developed and referenced in 1980 by Kawade, Y. J. Interferon Res. 1, 61-70 (1980). As far as I am aware it was created using purified mouse IFNB and not recombinant protein. However I do not have access to the original paper and cannot be sure of this. The first 22aa of the IFNB is the signal peptide region. I would not imagine the epitope of this antibody to be against this region and believe the peptide which you have found to be ideal. I believe that you could use ab24324 in that application confidently. If you would like to participate in the Abreview 100% discount program for Western blot I will gladly do that for you. I could also enroll you into this program for IF however each code needs to be associated with a different vial so in order to partake of two codes you will need to purchase two vials of ab24324. Alternatively I would like to encourage you to take advantage of our normal Abreview program for the testing in IF. It takes just a few minutes to leave a review and you can collect Abpoints which you may redeem for Abcam products or Amazon gift cards while at the same time sharing information about the product with your colleagues worldwide. More information may be at: https://www.abcam.com/index.html?pageconfig=resource&rid=10332 I hope that this information is helpful. Please let me know if you have any questions or there are other ways that Abcam may help you meet your research goals.

Answer

Thank you for your reply.

I do understand your concerns. It may be that you would want to run a western blot with this product to ensure that it is picking up IFN from your cells, it would them be possible to run a purified protein as control since we are unsure the hepatocytes would express. If this worked you could then stain with more confidence.

I hope this information is helpful to you. Please do not hesitate to contact us if you need any more advice or information.

Answer

Thank you for contacting us. We have tested this antibody in indirect ELISA and neutralization assays only. We haven’t tested this antibody in sELISA in combination with other antibodies so we will not be able to recommend the suitable ELISA pair for this product. Please be advised that sandwich ELISA procedures can be difficult to optimize and tested match pair antibodies should be used. This ensures the antibodies are detecting different epitopes on the target protein so they do not interfere with the other antibody binding. Therefore, we are unable to guarantee our antibodies in sandwich ELISA unless they have been specifically tested for sandwich ELISA. Please review antibody datasheets for information on tested applications. This antiobody has been used in sandwich ELISA as a capture antibody in 2 publications; González-Navajas JM et al. Interleukin 1 receptor signaling regulates DUBA expression and facilitates Toll-like receptor 9-driven antiinflammatory cytokine production. J Exp Med 207:2799-807 (2010). ELISA; Mouse. PubMed: 21115691 Cross JL et al. CD45 Regulates TLR-Induced Proinflammatory Cytokine and IFN-{beta} Secretion in Dendritic Cells. J Immunol 180:8020-9 (2008). ELISA; Mouse. PubMed: 18523265 I hope this information is helpful to you. Please do not hesitate to contact us if you need any more advice or information.
