Thank you for your enquiry and for taking your time to fill in the on-line Questionnaire and forwarding the images. Could we remind you that ab7206 is a polyclonal antibody and it may recognize different epitopes. Even the scanned Western blot image on our website shows non-specific bands. According to the Swiss-Prot datasheet, the expected band size is 10,73 kDa which is not that different from what the customer sees on the blot at around 13 kDa. There are disulfide sites along the peptide chain at positions 30 and 32 which may suggest some potential post-translational modifications. These may be good binding sites for other SH-containing molecules which can potentially create cross-links with the protein and alter the predicted molecular weight (although this is just prediction). Does the customer use pre-stained markers? If so, than the coupling of the chromophore to the polypeptide chains can actually affect the electrophoretic mobility in SDS-gel ELFO so it is not advisable to use the markers for quantitave studies (only for qualitative tools to determine approximate size). The antibody has been tested and characterized for Western blot application using rat brain lysate but rat spleen can also be used as good positive control. We hope this will help.
