Application Abreviews Notes
IHC-Fr Use at an assay dependent concentration.
ELISA Use at an assay dependent concentration. To detect hIP-10 by direct ELISA (using 100µl/well antibody solution) a concentration of at least 0.5µg/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with compatible secondary reagents, allows the detection of 0.2 - 0.4 ng/well of recombinant hIP-10.
Neutralising Use at an assay dependent concentration. To yield one-half maximal inhibition [ND50] of the biological activity of hIP-10 (100 ng/ml), a concentration of 2.0 - 3.0 µg/ml of this antibody is required.
WB Use at an assay dependent concentration. To detect hIP-10 by Western Blot analysis this antibody can be used at a concentration of 0.1 - 0.2 µg/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hIP-10 is 1.5 - 3.0 ng/lane, under either reducing or non-reducing conditions.
IHC-P Use at an assay dependent concentration.

  • ab9807 staining IP10 in Human carcinoid tissue sections by Immunohistochemistry (IHC-P - paraformaldehyde-fixed, paraffin-embedded sections). Antigen retrieval was by heat mediation in a pH 6.0 sodium citrate buffer. Samples were incubated with primary antibody (1ug/ml) overnight at 4°C. A HRP polymer detection system was used with a DAB chromogen.

Tissue underwent heat mediated antigen retrieval in sodium citrate buffer (pH 6.0). The primary antibody was used at 1.0 ug/ml and incubated with sample at 4°C overnight. A HRP-labeled polymer detection system was used with a DAB chromogen

    Tissue was fixed with paraformaldehyde, blocked with 5% serum for 1 hour at 24°C and permeabilized with Triton X-100. Samples were incubated with primary antibody (1/500 in PBS + 0.3% Triton X-100 + 1% BSA) for 3 hours at 24°C. An AlexaFluor®488-conjugated donkey anti-goat IgG polyclonal (1/1000) was used as the secondary antibody.

Ich habe die Immunogensequenz des ab9807 mit der IP10 Proteinsequenz von Maus für Sie verglichen. Leider stimmen beide Sequenzen nur zu 68% überein. Wir sehen es als wahrscheinlich an, dass ein Antikörper ein homologes Protein erkennt wenn die Übereinstimmung der homologen Sequenz mit der Immunogensequenz zu 85% übereinstimmt. Daher würden wir nicht erwarten dass der ab9807 das Maus IP10 erkennt.
Wenn Sie mir mitteilen für welche Anwendung genau Sie einen IP10 Antikörper suchen würde ich Ihnen gerne helfen und schauen ob einer unserer 14 anderen IP10 Antikörper für Sie in Frage kommt. Zwei Antikörper aller angebotenen IP10 Antikörper sind beispielsweise für Maus getestet.

Although we currently do not have the information about the isotype of ab9807 available, I have no doubt that it is a rabbit IgG. Nevertheless, I have contacted the lab to confirm this, and will update the datasheet once we have received the information.

As you can see from the Western blot image (and legend) on the datasheet, indeed any standard anti-rabbit IgG HRP-conjugated secondary (such as ab97080) can be used for detection.
