  • Function

    May mediate the control of various cellular processes by insulin. When phosphorylated by the insulin receptor binds specifically to various cellular proteins containing SH2 domains such as phosphatidylinositol 3-kinase p85 subunit or GRB2. Activates phosphatidylinositol 3-kinase when bound to the regulatory p85 subunit.

    Polymorphisms in IRS1 may be involved in the etiology of non-insulin-dependent diabetes mellitus (NIDDM) [MIM:125853].

  • Extracts of 293T cells transfected with wild-type human IRS1 and treated with 100 ng/mL TPA for 30 minutes were resolved by SDS-PAGE on a 10% Tris-glycine gel and transferred to PVDF.

Tissues were washed extensively in PBST and detection was performed using an HRP-conjugated secondary antibody followed by colorimetric detection using a DAB kit. Tissues were counterstained with hematoxylin and dehydrated with ethanol and xylene to prep for mounting.

Thank you for your enquiry and patience. I contacted the originator of this antibody to see if they could provide clarification regarding this. Phospho-IRS-1 (Ser616) in human corresponds to Ser612 of mouse IRS-1. Also, the human, rat and mouse sequences are 100% homologous (identical, even) for this immunogenic sequence, but we can't confirm that the antibody will recognize all 3 species, since we've not tested them (recognition is expected, but not tested/confirmed).
