  • Species reactivity
    Reacts with: Human
Predicted to work with: Mouse, Rat
  • Immunogen

Synthetic peptide derived from a region of IRS 1 that contains serine 616. The sequence is conserved in human, mouse, and rat.

  • Positive control
    • CHO cell line transfected with a vector encoding the human insulin receptor (CHO-T) and transiently transfected with a vector encoding IRS 1, stimulated with a phorbol ester, TPA.

Our Abpromise guarantee covers the use of ab4776 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
WB Use a concentration of 0.1 - 1 µg/ml. Predicted molecular weight: 165 kDa.

  • Function
May mediate the control of various cellular processes by insulin. When phosphorylated by the insulin receptor binds specifically to various cellular proteins containing SH2 domains such as phosphatidylinositol 3-kinase p85 subunit or GRB2. Activates phosphatidylinositol 3-kinase when bound to the regulatory p85 subunit.
  • Involvement in disease
Polymorphisms in IRS1 may be involved in the etiology of non-insulin-dependent diabetes mellitus (NIDDM) [MIM:125853].
  • Sequence similarities
    Contains 1 IRS-type PTB domain.
    Contains 1 PH domain.
  • Post-translational
Serine phosphorylation of IRS1 is a mechanism for insulin resistance. Ser-312 phosphorylation inhibits insulin action through disruption of IRS1 interaction with the insulin receptor.
Phosphorylation of Tyr-896 is required for GRB2-binding.

  • Extracts prepared from CHO-T cells mock transfected (Mock – lanes 1, 4), transiently transfected with wild-type human IRS 1 (WT – lanes 2, 5), and transiently transfected with mutant human IRS 1 (S616A – lanes 3, 6) and treated with TPA were resolved by SDS-PAGE on a 10% Tris-glycine gel and transferred to nitrocellulose. After blocking non-specific sites with a 5% BSA-TBST buffer overnight at 4oC, the membranes were incubated with 0.50 µg/mL ab4776 antibody (lanes 1-3), or an anti-IRS 1 pan antibody (lanes 4-6). After washing, membranes were incubated with goat F(ab’)2 anti-rabbit IgG alkaline phosphatase. Signals were detected using the Tropix WesternStar method. The data show that the ab4776 antibody yields a signal on only the wild-type IRS 1 extract, not the mock or mutant IRS 1 extracts, thereby demonstrating the specificity of the antibody to this site. The data also show that the overall level of IRS 1 does not change between the wild-type and th

Customer reviews and Q&As

Thank you for your enquiry and patience. I contacted the originator of this antibody to see if they could provide clarification regarding this. Phospho-IRS-1 (Ser616) in human corresponds to Ser612 of mouse IRS-1. Also, the human, rat and mouse sequences are 100% homologous (identical, even) for this immunogenic sequence, but we can't confirm that the antibody will recognize all 3 species, since we've not tested them (recognition is expected, but not tested/confirmed). If you have any additional questions, please contact us again.
