    IHC image of Islet 1 staining in a section of formalin fixed, paraffin embedded mouse embryo E12, performed on a Leica Bond™ system using the standard protocol B. The section was then incubated with ab20670, 0.5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen.     For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

    ab20670 2µg/ml staining ISLET1 in human pancreas using an automated system (DAKO Autostainer Plus). Using this protocol there is strong nuclear and weak cytoplasmic staining primarily in the pancreatic islet.
    Sections were rehydrated and antigen retrieved with the Dako 3 in 1 AR buffer EDTA pH 9.0 in a DAKO PT Link. Slides were peroxidase blocked in 3% H2O2 in methanol for 10 mins. They were then blocked with Dako Protein block for 10 minutes (containing casein 0.25% in PBS) then incubated with primary antibody for 20 min and detected with Dako Envision Flex amplification kit for 30 minutes. Colorimetric detection was completed with Diaminobenzidine for 5 minutes. Slides were counterstained with Haematoxylin and coverslipped under DePeX. Please note that, for manual staining, optimization of primary antibody concentration and incubation time is recommended. Signal amplification may be required.

    Dorsal root ganglion explants were dissected from 16 day-old rat embryos and cultured for 6 hours in vitro with Neurobasal Medium containing B27 supplement.     

    Nuclei stained positive for anti-Islet 1 antibody ab20670 at 2µg/ml. As would be expected, not all cells in this preparation were Islet 1-positive. Pre-incubation of ab20670 with the immunizing peptide ab21996 resulted in complete blocking of the antibody.

The tissue was fixed with paraformaldehyde followed by blocking with 5% serum for 1 hour at 25°C. The tissue was incubated with ab20670 in PBST for 12 hours at 4°C. A polyclonal donkey anti-rabbit Alexa Fluor® 594 secondary antibody was used at 1/1000 dilution.

Question

Hi,
Thank for your tips- it has worked really well at 1:1000- very clean.
The only thing is that I am staining large neurospheres adn the tween
0.2% permealized into the center- I guess its too weak. Is there
anything else you could suggest for this?

Reviewing this case, I would like to offer some suggestions to help optimise the results. I would also appreciate if you can confirm some further details:

1. I would appreciate if you are able to provide an image which would help me to assess the results.

2. Could you confirm the wash steps? We recommend to wash 3 or 4 times at each appropriate wash step using PBS containing 0.2% Tween. This will help to wash away any excess antibody and also keep the cells permeabilized.

3. I agree that reducing antibody concentration would be beneficial to try. Try 1:1000.

4. Try reducing the secondary antibody concentration aswell.

Question

Inquiry: Hi, I have been trying to stain an in vitro culture of human neurons and I am getting very high amounts of background. I do see higher nuclear levels but this is very hard to resolve as the antibody appears to stain the cytoplasm and membrane! The unspecific staining is very bright. I am using this at the recommended dilutions found on this website and I know the problem is not the secondary antibody as this is working fine in combination with other primary antibodies. I also know the cells that I fixed were ok as the protein I co-stained showed a very specific pattern. I was wondering if you would be able to tell me if this lot. of antibody has been characterized? I would like to try another batch or an alternative antibody. In detail the cells were fixed in 4% paraformaldehyde, permealized with 0.1% tritonX in PBS. They were then blocked in 10% goat serum for an hour and finally incubated with the primary antibodies overnight at 4C, in 1% goat serum in PBS/TritonX at a 1:400 concentration. Cells were then washed with PBS and incubated with the secondary for 2 hours at room temperature. Secondary used was an alexafluor goat anti-rabbit 488 at 1:500. Cheers

I have initially used this antibody from a colleague in my IHC-P experiment and it worked great. this was lot GR26251. Then I ordered my own vial, GR43585, but this lot did not work on the same sections. I see faint specific staining in the nucleus but high background in the nucleus as well.
