    Cell line: K562 (Human Chronic Myelogenous Leukemia).
    Growth media: RPMI and 10% NCS (Newborn calf serum).

     

    K562 cell lysate was prepared by homogenization in modified RIPA buffer (150 mM sodium chloride, 50 mM Tris-HCl, pH 7.4, 1 mM ethylenediaminetetraacetic acid, 1 mM phenylmethylsulfonyl flouride, 1% Triton X-100, 1% sodium deoxycholic acid, 0.1% sodium dodecylsulfate, 5 µg/ml of aprotinin, 5 µg/ml of leupeptin). Cell debris was removed by centrifugation. Protein concentration was determined with Bio-Rad protein assay. The cell lysate was boiled for 5 min in 1 x SDS sample buffer (0.045 M Tris-HCl pH 6.8, 10% glycerol, 1% sodium dodecylsulfate, 0.01% bromophenol blue), containing 0.05 M DTT.

  • Background

    K-562 is an erythroleukemia cell line derived from a chronic myeloid leukemia patient in blast crisis. Recent studies have shown the K562 blasts are multipotential, hematopoietic malignant cells that spontaneously differentiate into recognisable progenitors of the erythrocyte, granulocyte and monocytic series. Cells are also used for haemoglobin synthesis, antitumour testing, tumorigenicity, differentiation, pharmacodynamics, cloning, cytotoxicity, cell biology, cellular effects of hyperthermia, and natural killer assays.

We are currently doing western blot analysis using "Anti-FNDC5 antibody-Carboxyterminal end (ab93373)" from abcam. As its datasheet mentions, K562 cell line lysate was used as positive control, which we are planning to orderas soon as possible. However, we are not sure whether we should order "K562 whole cell lysate-tumor cell line (ab29306)" or "k562 whole cell lysate (ab7911)" for this purpose. Moreover, could you please kindly email me the western blot condition when using this lysate?

Thank you for contacting us.

The lysates ab29306 and ab7911 were prepared using slightly different techniques, but will give equivalent results in WB. Either of these lysates will be guaranteed for use as a positive control with ab93373. We recommendloading 20ug of lysate per lane. Ab29306should be boiled for 5 minutes prior to loading in thegel, but ab7911 has already beenboiled.I hope this helps, please let me know if you need any additional information or assistance.

The lysate is vialed in terms of protein concentration, so unfortunately the lab does not have any information about the cell concentration.
