Our Abpromise guarantee covers the use of ab70183 in the following tested applications.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
|IHC-P||Use a concentration of 5 µg/ml. Perform heat mediated antigen retrieval with Tris/EDTA buffer pH 9.0 before commencing with IHC staining protocol.|
|WB||1/500 - 1/1000. Detects a band of approximately 71 kDa (predicted molecular weight: 71 kDa).|
|ICC/IF||1/100 - 1/500.|
|IP||Use a concentration of 5 µg/ml.|
FunctionComponent of the HBO1 complex which has a histone H4-specific acetyltransferase activity, a reduced activity toward histone H3 and is responsible for the bulk of histone H4 acetylation in vivo. Through chromatin acetylation it may regulate DNA replication and act as a coactivator of TP53-dependent transcription. Specifically represses AR-mediated transcription.
Tissue specificityUbiquitously expressed, with highest levels in testis.
Sequence similaritiesBelongs to the MYST (SAS/MOZ) family.
Contains 1 C2HC-type zinc finger.
DomainThe C2HC-type zinc finger is required for interaction with MCM2 and ORC1L.
The N-terminus is involved in transcriptional repression, while the C-terminus mediates AR-interaction.
modificationsPhosphorylated upon DNA damage, probably by ATM or ATR.
Cellular localizationNucleus > nucleoplasm.

IHC image of ab70183 staining in human breast carcinoma formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with EDTA (pH9, epitope retrieval solution 2) for 20 mins. The section was then incubated with ab70183, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.
For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.
KAT7 / Hbo1 / MYST2 was immunoprecipitated using 0.5mg Jurkat whole cell extract, 5µg of Rabbit polyclonal to KAT7 / Hbo1 / MYST2 and 50µl of protein G magnetic beads (+). No antibody was added to the control (-).
The antibody was incubated under agitation with Protein G beads for 10min, Jurkat whole cell extract lysate diluted in RIPA buffer was added to each sample and incubated for a further 10min under agitation.
Proteins were eluted by addition of 40µl SDS loading buffer and incubated for 10min at 70oC; 10µl of each sample was separated on a SDS PAGE gel, transferred to a nitrocellulose membrane, blocked with 5% BSA and probed with ab70183.
Secondary: Mouse monoclonal [SB62a] Secondary Antibody to Rabbit IgG light chain (HRP) (ab99697).
Band: 71kDa; KAT7 / Hbo1 / MYST2; non specific bands - 80kDa: We are unsure as to the identity of this extra band.
