    Histone demethylase that demethylates 'Lys-4' and 'Lys-36' of histone H3, thereby playing a central role in histone code. Preferentially demethylates trimethylated H3 'Lys-4' and dimethylated H3 'Lys-36' residue while it has weak or no activity for mono- and tri-methylated H3 'Lys-36'. Preferentially binds the transcribed region of ribosomal RNA and represses the transcription of ribosomal RNA genes which inhibits cell growth and proliferation. May also serve as a substrate-recognition component of the SCF (SKP1-CUL1-F-box protein)-type E3 ubiquitin ligase complex.

While we have not yet tested this product in that application I was hoping that I might help you to achieve signal with this product. In reviewing your protocol I noticed that paraformaldehyde was used to fix your tissue. Because of the nature of this fixative, use of Pfa will often mask the epitope sites which the antibody will recognize. In order to keep these sites available we recommend use of methanol or acetone to fix tissue. If this is not possible I would recommend use of an antigen retrieval step. An effective antigen retrieval protocol for frozen tissues is to heat Citrate buffer pH9.0 to +80C in a water bath. Before permeablizing your sections, place then within the heated buffer leaving them for 30 minutes. Follow this with a tap water rinse and then a DI water rinse.
