    Histone demethylase that specifically demethylates 'Lys-27' of histone H3, thereby playing a central role in histone code. Demethylates trimethylated and dimethylated but not monomethylated H3 'Lys-27'. Plays a central role in regulation of posterior development, by regulating HOX gene expression. Demethylation of 'Lys-27' of histone H3 is concomitent with methylation of 'Lys-4' of histone H3, and regulates the recruitment of the PRC1 complex and monoubiquitination of histone H2A.

Respecto a si el anticuerpo puede o no estar detectando las isoformas correspondientes a pesos moleculares alrededor de 100KDa, no tengo constancia de que así sea. De todas maneras puedo contactar con el laboratorio que seguramente dispongan de más información acerca de su especificidad.

En cualquier caso, dado que incluso el control positivo os da una banda a 100KDa, yo aconsejaría cambiar el protocolo de lisis de las células. Indicáis en el protocolo que se ha usado un buffer consistente en Tris HCl, NaCl y Triton X100. Este buffer suele utilizarse en muestras que no requieren desnaturalización.

El anticuerpo ab84190 reconoce la proteína en estado reducido y desnaturalizado, por lo que yo sugeriría cambiar a un buffer como RIPA, recomendado en estos casos.

La datasheet del anticuerpo también aconseja usar un agente de bloqueo especifico, 5% de leche en polvo en 0.05% PBS-T.

Y como ultimo chequeo para descartar posibles causas de bandas inespecíficas, os recomiendo también que llevéis a cabo un control no primario para averiguar si el anticuerpo secundario puede dar lugar a bandas de este tipo.

Os propongo que probéis estas variaciones en el protocolo y en caso de seguir sin obtener el resultado esperado me volváis a contactar. Si el anticuerpo resultara haber salido defectuoso, al estar en garantía, os mandaríamos un vial nuevo gratuito o un reembolso en compensación.
