                  The advantages of high-throughput FirePlex-384 and FirePlex-1536 immunoassays

                  ​Accelerate next discovery with a fast, automated workflow

                  FirePlex immunoassays use the FirePlex particle technology and validated antibody pairs to deliver rapid, robust, and sensitive high-throughput multiplex screening without the need for dedicated instrumentation. Using a high content imager, you can analyze up to:

                  • 10 analytes/well with FirePlex-384, < 20 min scan per plate
                  • 5 analytes/well with FirePlex-1536, < 1 hour scan per plate

                  FirePlex-384 and ​​FirePlex-1536 are ideally suited for high-throughput automated workflows (Figure 1). You can use any compatible liquid handling robot for buffer and sample dispensing into wells to simplify preparation and eliminate the ergonomic load on lab personnel caused by repeat pipetting.

                  Figure 1. The assay workflow. To capture analytes onto FirePlex particles, biological samples are added to a 384-well imaging plate and incubated with the FirePlex-384 Reagent Mix overnight. Subsequently, an Imaging Dye is added, and plates are scanned on high-content imagers. The FirePlex Analysis Workbench software generates standard curves and quantifies analytes of interest directly from image files.

                  ​Gather more insightful data for drug development applications

                  High-throughput FirePlex immunoassays can be applied at all stages of drug development to inform critical decision points:

                  • Discovery projects screening large compound libraries
                  • Lead optimization, safety assessment, and toxicity screening in large sample sets
                  • Clinical screening for disease identification, patient stratification, or therapy response in large patient cohorts

                  By providing a new, more effective biomarker analysis workflow (Figure1), FirePlex immunoassays enable data-driven R&D to guide the most effective strategies and minimize the risk of late-stage failures. ​

                  Scale-up without sacrificing performance

                  Our FirePlex immunoassays offer high reproducibility and sensitivity, so you can achieve high throughput without sacrificing assay performance or data quality.

                  • 1-100 pg/mL sensitivity (analyte dependent)
                  • 2.5-3 logs dynamic range
                  • Only 6.5 µL biofluid input for 384-well plate (plasma, serum, or cell culture supernatant); 2.5 µL biofluid input for 1536-well plate (Cell culture supernatant)
                  • <15% intra-plate CVs
                  • The use of stringent production and QC processes ensures lot-to-lot consistency
                  • 70-130% sample recovery
                  • Re-read plates up to a week later, as many times as needed

                  ​Be flexible with analytes

                  • Top 50 key cytokines and subsets of critical organ toxicity markers

                  Simple data collection on high content imagers

                  FirePlex-384 and FirePlex-1536 assays are compatible with high content imagers from various prominent manufacturers, including Molecular Devices, Perkin Elmer, GE, Yokogawa, Agilent, and Thermo. Check the required characteristics in Figure 2 below.
