ICC/IF Use a concentration of 5 µg/ml.
WB Use a concentration of 1.25 µg/ml. Detects a band of approximately >70 kDa (predicted molecular weight: 288 kDa). Good results were obtained when blocked with 5% non-fat dry milk in 0.05% PBS-T.
IHC-P Use a concentration of 4 - 8 µg/ml.

    Histone methyltransferase that methylates 'Lys-36' of histone H3. H3 'Lys-36' methylation represents a specific tag for epigenetic transcriptional activation. Probably plays a role in chromatin structure modulation during elongation via its interaction with hyperphosphorylated POLR2A. Binds DNA at promoters. May also act as a transcription activator that binds to promoters. Binds to the promoters of adenovirus 12 E1A gene in case of infection, possibly leading to regulate its expression.

    why is the actual band size different from the predicted?

  • Immunohistochemical analysis of paraffin-embedded human placenta using ab69836 at 4 µg/ml. Positively labelled trophoblast cells are indicated with arrows.
  • ICC/IF image of ab69836 stained HepG2 cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab69836, 5µg/ml) overnight at +4°C. The secondary antibody (green) was Alexa Fluor® 488 goat anti-rabbit IgG (H+L) used at a 1/1000 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

Thank you for your inquiry.

Most antibodies will work fine with either BSA or milk.

When testing our antibodies, our lab uses 5% BSA as a blocking reagent, so I recommend this.

Some antibodies give stronger, more specific signals on blots blocked with BSA instead of milk, so doing this may improve the results you are seeing, and reduce the non-specific bands.

An example of the above is the western blot obtained with the Abcam GAPDH antibody ab9385 : https://www.abcam.com/ab9385 .

Unfortunately, this has to be tested experimentally according to your experimental set-up. But I suggest top start with BSA and only change the blocking agent when there is need.
