Beta-Glucosidase Assay Kit (ab272521) is a simple, direct and automation-ready procedure for measuring β-glucosidase activity. beta-Glucosidase Assay Kit is designed to measure β-glucosidase activity directly in biological samples without pretreatment. The improved method utilizes p-nitrophenyl-β-D-glucopyranoside that is hydrolyzed specifically by β-glucosidase into a yellow colored product (maximal absorbance at 405nm). The rate of the reaction is directly proportional to the enzyme activity.

High sensitivity and wide linear range. Use 20 μL sample. The detection limit is 2 U/L, linear up to 250 U/L.

Homogeneous and simple procedure. Simple "mix-and-measure" procedure allows reliable quantitation of β-glucosidase activity within 20 minutes.

Robust and amenable to HTS. All reagents are compatible with high-throughput liquid handling instruments.

Beta-glucosidase also known as β-glucosidase or GCase is an enzyme that catalyzes the hydrolysis of β-glucosidic bonds in β-glucosides releasing glucose. This enzyme has a molecular mass of around 55 kDa and occurs in various organisms including plants microorganisms and humans. In humans beta-glucosidase is expressed in tissues such as the spleen liver and kidney. The protein plays a mechanical role in cleaving glucose from different glycosides which is important for various metabolic processes.

Beta-glucosidase functions by aiding the degradation of complex carbohydrates facilitating the release of glucose for cellular energy use and material recycling. This enzyme is not typically part of a large molecular complex but operates independently to fulfill its function. Its activity is essential for breaking down plant-derived compounds and is significant in the metabolic processes that contribute to energy production and detoxification of harmful glucosides.

Pathways

Beta-glucosidase participates in the glycosphingolipid metabolism and cellulose degradation pathways. In glycosphingolipid metabolism it is involved in the lysosomal pathway where it breaks down glucosylceramide into ceramide and glucose. This activity impacts protein networks that include lysosomal enzymes. In cellulose degradation beta-glucosidase acts as a final step converting cellobiose into two glucose molecules linking it to other carbohydrate-active enzymes like cellulase.

Beta-glucosidase deficiency is connected to Gaucher's disease a lysosomal storage disorder where glucosylceramide accumulates due to insufficient breakdown. This enzyme's dysfunction also indirectly impacts proteins involved in lysosomal function contributing to disease pathology. Additionally beta-glucosidase may associate with Parkinson's disease through its role in cellular metabolism and the energy dysfunction observed in neurodegenerative conditions. The protein interacts with other metabolic and lysosomal enzymes indicating its broader involvement in health and disease contexts.
